摘 要:目的:耳柄蒲儿根萃取物的抗氧化活性研究。方法:采用水萃取法、大孔树脂AB8吸附和50%乙醇溶剂洗脱方法获取耳柄蒲儿根萃取物;以没食子酸为标准品,检测耳柄蒲儿根萃取物多酚含量;通过总抗氧化能力(ABTS试剂盒法),1,1-二苯基-2-三硝基苯肼(DPPH)自由基的清除能力,一氧化氮(NO)清除能力以及对乙酰氨基酚(APAP)诱导的人肝肿瘤细胞株HepG2氧化损伤测定,4项指标测定耳柄蒲儿根萃取物的抗氧化损伤作用能力。结果:测得该萃取物总酚含量为53.21μg GAE/mg,耳柄蒲儿根萃取物的总抗氧化活性为5.359 mmol/g,对DPPH自由基的半数抑制率浓度(IC_(50)值)为0.52±0.02 mg/mL,在1 mg/mL浓度时对NO自由基的相对清除率为6.68%,在浓度为0.08 mg/mL时对人肝肿瘤细胞株HepG2受到的APAP诱导的氧化损伤具有一定修复作用。结论:上述实验结果初步证实耳柄蒲儿根萃取物具有良好的抗氧化能力。Objective:To investigate the antioxidant activity of Sinosenecio euosmus(Handel-Mazzetti)B.Nordenstam.Methods:Antioxidant activities were determined using three assays.Total antioxidant capacity assay(2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid(ABTS)),1,1-Diphenyl-2-picrylhydrazyl(DPPH)radical scavenging assay,and the model of hydrogen peroxide induced oxidative stress of liver cancer cells HepG2.Results:The results showed that its total antioxidant capacity was 5.359 mmol/g and its concentration providing 50% inhibition(IC50)was 0.52 mg/mL in the DPPH test and 0.074 mg/mL in superoxide anion radical scavenging test.The relative scavenging rate of NO was 6.68%at the concentration of 1 mg/mL.The results also showed that S.euosmus(Handel-Mazzetti)B.Nordenstam extracts could significantly protect HepG2 from APAP induced oxidative stress at the concentration of 0.08 mg/mL.Conclusion:These results demonstrated that S.euosmus(Handel-Mazzetti)B.Nordenstam has good antioxidant capacity.
