摘  要:试验旨在探究饲粮添加茶多酚对乳鸽十二指肠形态发育、抗氧化功能及紧密连接蛋白基因mRNA表达量的影响,为茶多酚作为功能性饲料添加剂在养鸽生产中的合理应用提供理论依据。选取初始体重一致、健康1日龄仔鸽240只,随机分为4组,每组6个重复,每个重复10只仔鸽。对照组饲喂基础饲粮,茶多酚低、中、高剂量组分别在基础饲粮中添加100、200和400 mg·kg^(-1)茶多酚,试验期28 d。试验结束后,采集乳鸽十二指肠组织进行肠道形态发育、抗氧化功能以及紧密连接蛋白基因表达测定。结果表明,与对照组相比,中剂量茶多酚极显著提高乳鸽十二指肠绒毛高度(VH)、隐窝深度(CD)、提高绒隐比(VH/CD)(P<0.01),高剂量茶多酚极显著提高乳鸽十二指肠绒毛高度和隐窝深度(P<0.01);低剂量茶多酚极显著降低乳鸽十二指肠丙二醛(MDA)含量(P<0.01),中剂量茶多酚极显著降低乳鸽十二指肠MDA含量和乳酸脱氢酶(LDH)活性(P<0.01),高剂量茶多酚极显著提高乳鸽十二指肠总抗氧化能力(T-AOC)、谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)活性,降低MDA含量和LDH活性(P<0.01),3个剂量茶多酚均极显著提高SOD2基因mRNA表达量(P<0.01),高剂量茶多酚还极显著提高过氧化氢酶(CAT)基因mRNA表达量(P<0.01);此外,低剂量茶多酚极显著提高Occludin、降低Claudin-2 mRNA表达量(P<0.01),中剂量茶多酚极显著提高Occludin和ZO-1并降低Claudin-2 mRNA表达量,高剂量茶多酚极显著提高Occludin和ZO-1基因mRNA表达量,显著提高Claudin-3基因mRNA表达量(P<0.05)并极显著降低Claudin-2 mRNA表达量(P<0.01)。综上所述,饲粮添加茶多酚可以通过改善乳鸽十二指肠肠道发育、提高肠道抗氧化能力和调节肠道紧密连接蛋白相关基因mRNA表达量进而维持乳鸽肠道健康。The aim of this study was to investigate the effects of tea polyphenols(TP) on duodenal morphological development, antioxidant function and mRNA expression levels of intestinal tight junction protein genes in squabs to provide a theoretical basis for the rational application of TP as a functional feed additive in the production of pigeons. First, young pigeons(240 pairs, 1 day old,healthy, similar body weight) were randomly divided into four groups(control group fed on basal feed,100 mg · kg^(-1) TP treatment group, 200 mg · kg^(-1) TP treatment group, and 400 mg · kg^(-1) TP treatment group) with six replicates per group and ten pigeons per replicate(the content of TP≥ 98%). After a 28 days feeding period, pigeon duodenum was collected for the detection of duodenum morphological development, antioxidant function and mRNA expression levels of intestinal tight junction protein genes. The results showed that when compared with squabs in the control group, feeds with TP could promote squabs’ duodenum development. Here, feeds with medium amount(200 mg · kg^(-1)) of TP could increase duodenum intestinal villus height(VH), crypt depth(CD), velvet to hidden ratio(VH/CD)extremely significantly(P<0.01) among squabs. Further, high amount of 400 mg · kg^(-1) TP could increase duodenum VH and VH/CD(P<0.01) significantly among squabs. Besides, the results had shown that feeds added with TP could elevate intestinal antioxidant functions in TP-treatment groups compared with the control ones. Specifically, lower amount of TP(100 mg · kg^(-1)) could decrease duodenum malondialdehyde(MDA) content extremely significantly(P<0.01), while medium amount of TP could reduce MDA content and lactic dehydrogenase(LDH) activities extremely significantly(P<0.01). Also,feeds with high amount of TP could lead to increasing in the total antioxidant capacity(T-AOC),glutathione peroxidase(GSH-Px), superoxide dimutase(SOD) activities and reduction in MDA contents and LDH activities extremely significantly(P<0.01).
