摘 要:为了分离筛选出一种大麦草青贮优良菌株并验证其生产性能,从大麦草中筛选出产酸能力较强的菌株,进行生长速度测定与回接青贮实验,复筛选出最优菌株,进行16srDNA鉴定,鉴定菌株与非大麦草青贮专用菌株进行对比实验,研究分离菌株在大麦草青贮中的应用效果。结果表明,由大麦牧草中分离获得11株产酸杆菌,经过产酸能力检测,确定了3株产酸量差异显著(P<0.05)的菌株,回接青贮实验,筛选出最适菌株;与非大麦草青贮专用菌株比较结果显示,分离菌菌株产酸速度优于对比菌株,差异显著(P<0.05)。经16srDNA鉴定,该菌株为植物乳杆菌(Lactobacillus plantarum)。由大麦草中分离纯化的植物乳杆菌生长速度和产酸能力优于野生及市售非专用菌株,可作为大麦草青贮专用菌株,在大麦草青贮生产中应用。To isolate and screen an excellent strain for barley grass silage and to verify its production performance,bacterial strain with relatively strong acid production from wheat straw hood were selected to test growth rate and back-join silage experiment,and screen out the best strains,the best strains were screened out and identified by 16s rDNA identification.The identified strains were compared with non barley grass silage special strains to detect the application effect of the isolated strains.The results showed that 11 strains of acid producing bacteria were isolated from barley forage.3 strains with significant difference in acid production(P<0.05)were identified in the acid production ability test.The most suitable strains were screened out in the back-joining silage experiment.Compared with non barley forage silage special strains,the acid production rate of isolated strains was significantly better than that of two non special strains(P<0.05).The strain was identified as Lactobacillus plantarum by 16srDNA.The growth rate and acid production ability of Lactobacillus plantarum isolated and purified from barley grass were better than those of wild and commercial non special strains.The strain can be used in barley grass silage production as a special strain.
