摘 要:铁死亡是一种新型的铁依赖性脂质过氧化物损伤诱导的调节性细胞死亡形式,其在形态学、遗传学和分子生物学等方面都不同于凋亡、自噬和坏死等其他细胞死亡方式。铁死亡与肿瘤之间关系极为密切,目前已有大量研究设计和开发基于铁死亡的抗癌药物,而铁死亡有望成为一种新颖的肿瘤治疗方法。本文首先总结了铁死亡的发生机制以及参与铁死亡调控的细胞器,以阐明铁死亡特征及其在肿瘤治疗方面的可行性,然后介绍了铁死亡在化学、材料学、癌症生物学及中药学交叉领域中治疗肿瘤的最新应用进展。Ferroptosis is a newly discovered form of regulated cell death induced by iron-dependent lipid peroxide injury,which is different from other cell death modalities of apoptosis,autophagy and necrosis in the aspect of morphology,genetics and molecular biology. Ferroptosis is closely related to tumor,and there have been a variety of research to design and develop anti-cancer drugs based on ferroptosis induction,and ferroptosis is expected to be a novel tumor treatment method. The mechanism of ferroptosis and the organelles involved in ferroptosis are summarized firstly to highlight the feasibility and characteristics of triggering ferroptosis for cancer therapy.Meanwhile,the latest advances in the application of ferroptosis-inducing agents at the intersection of chemistry,materials science,cancer biology and Chinese herbs for the cancer therapy was introduced.
