摘  要:目的探讨plakoglobin(PG)和PPARγ蛋白在三阴性乳腺癌(triple-negativebreast cancer,TNBC)组织中的表达及临床意义。方法应用免疫组织化学方法和Western blot方法检测TNBC组织中PG和PPARγ蛋白的表达情况,并分析两者与TNBC临床病理特征及预后的关系。结果和癌旁正常组织比较,PG和PPARγ在TNBC组织中的表达水平明显降低(P<0.05);PG和PPARγ的表达随着淋巴结转移和临床分期的增高而降低(P<0.05);PG和PPARγ的表达呈正相关(r=0.485,P=0.007)。Kaplan-Meier生存分析显示淋巴结转移、高级别TNM分期、PG和PPARγ的阴性表达与TNBC患者生存期缩短相关。Cox回归分析提示高级别TNM分期以及PG和PPARγ双阴性表达是TNBC患者预后差的主要危险因素。结论 PG和PPARγ的表达抑制可能协同促进TNBC的恶性进展,两者可作为TNBC患者治疗的靶标及预后评估的重要参考依据。Objective The goal of this study was to explore the expressions of plakoglobin(PG) and PPARγ in triple-negative breast cancer(TNBC) and their clinical significance. Methods Immunohistochemistry and Western blot methods were used to determine the levels of PG and PPARγ expression in TNBC tissues, and the correlation of PG and PPARγ expression with clinicopathological characteristics and prognosis of the patients with TNBC was analyzed.Results Compared with cancer adjacent normal tissues, the expression levels of PG and PPARγ were significantly decreased in TNBC tissues(P<0.05). The decreased expressions of PG and PPARγ were correlated with lymph node metastasis and advanced TNM stage(P<0.05). The expression of PG was positively correlated with PPARγ expression in TNBC tissues(r=0.485, P=0.007). Kaplan-Meier analysis revealed that lymph node metastasis, advanced TNM stage as well as negative expression of PG or PPARγ were correlated with shorter survival time in patients with TNBC. Cox regression analysis confirmed that advanced TNM stage and double negative expression of PG and PPARγ might be important on the point of unfavorable prognostic factors for TNBC patients. Conclusion The inhibition of PG and PPARγ expression may collaboratively promote the malignant progression of TNBC, and both of them could serve as the important targets for the treatment of TNBC patients, as well as the significant indicators for the prognosis evaluation of TNBC patients.
