摘  要:为研究蒙古马耐粗饲料的优良特点,试验以蒙古马盲肠内容物为研究对象,旨在筛选、分离出高效的蒙古马源纤维素分解菌株,并对分离菌株进行鉴定和酶学特性分析。采用羧甲基纤维素平板法和摇瓶发酵法分离菌株并鉴定菌株类型。最后,通过测定纤维素酶活力分析菌株在不同pH、温度、碳源和氮源条件下的酶学特性。研究从蒙古马盲肠内容物中分离筛选到具有较高降解纤维素能力的解淀粉芽孢杆菌H3(Bacillus amyloliquefaciens H3)。菌株H3分泌纤维素酶的最适p H为4.8,在p H 3.0~8.0范围内,其酶活力相对稳定;在温度为65℃,酶活力最高,当温度为40~50℃时,该酶活力维持在80%左右,稳定性强。此外,H3发酵所需的最佳碳源为玉米粉,最佳氮源为2%的蛋白胨+酵母粉复合物(1:1),它具有相对较高的纤维素酶活力,有深入开发的潜力。In order to study the roughage tolerance characteristics of Mongolian horses,this experiment took Mongolian horse’s cecum content as the research object,and aimed to isolate high-efficiency cellulolytic strains derived from Mongolian horse.In addition,the isolated strains were identified and analyzed for enzymatic characteristics.The carboxymethyl cellulose plate method and shake flask fermentation method were used to isolate the strains and identify the strain type.Finally,by measuring the cellulase activity,the enzyme properties of the strains under different pH,temperature,carbon source and nitrogen source conditions were analyzed.In this study,Bacillus amyloliquefaciens H3 was isolated and screened from the contents of Mongolian horses cecun.The optimal pH for cellulase secretion by strain H3 was 4.8.In the range of pH 3.0-8.0,the enzyme activity was relatively stable;at temperature of 65 ℃,the enzyme activity was the highest,and when the temperature was between 40-50 ℃,the enzyme activity remains at around 80%with strong stability.Furthermore,the best carbon source for H3 fermentation was corn flour,and the best nitrogen source was 2% peptone+yeast powder complex(1:1).The H3 fermentation had relatively high cellulase activity and promisiing development potential.
