摘 要:以乙酸、乙酸-双氧水两步预处理后的杨木残渣为底物,探讨化学助剂的种类和浓度对纤维素酶水解的影响,并以纤维素酶水解液中单糖为碳源,对枯草芽孢杆菌进行发酵培养,探究表面活性剂种类和剂量对其发酵过程的影响。结果表明:额外添加1000nkat/g的β-葡萄糖苷酶,杨木葡萄糖和木糖得率分别提高到74.35%和95.43%。添加吐温-80、PEG-6000、木质素磺酸钠对酶水解均有促进作用,且PEG-6000促进效果更为显著。不同表面活性剂对发酵有不同影响,吐温-80和PEG-6000在一定程度上抑制菌株生长,而木质素磺酸钠对发酵有促进作用,添加量为1g/L时活菌数提高到2.10×10^(9)CFU/mL。研究结果为木质纤维素的生物转化益生菌提供了科学依据。Taking the poplar residue with acetic acid and acetic acid hydrogen peroxide as the substrate,the effects of the types and concentrations of chemical additives on cellulase hydrolysis were studied,and Bacillus subtilis was cultured with monosaccharide in cellulase hydrolysate as carbon source to explore the effects of the types and doses of surfactants on its fermentation process.The results showed that the yield of glucose and xylose were increased to 74.35%and 95.43%respectively by adding 1000 nkat/gβ-glucosidase.Adding Tween-80,PEG-6000,and sodium lignosulfonate all had a promoting effect on enzymatic hydrolysis,but the promotion effect of PEG-6000 was more significant.The addition of Tween-80 or PEG-6000 could inhibit the growth of the strain in varying degrees.The difference is that sodium lignosulfonate can promote the fermentation of Bacillus subtilis.1 g/L sodium lignosulfonate could increase the number of viable Bacillus subtilis to 2.10×10^(9) CFU/mL.The study laid a theoretical foundation for the high-value utilization of renewable energy such as lignocellulose.
