摘 要:考察了葡萄糖和木糖对蛹虫草深层液体发酵的影响,并研究了蛹虫草利用玉米芯发酵生产虫草素的可行性。结果表明,当以葡萄糖或木糖为唯一碳源时,最适碳源为30 g/L葡萄糖或20 g/L木糖,虫草素产量分别达到(144.63±19.48)mg/L和(50.61±0.66)mg/L。建立了葡萄糖-木糖两阶段发酵,虫草素生产强度达到(3.55±0.09)mg/(L·d),较20 g/L木糖发酵体系提高了40.32%。通过优化玉米芯酶解条件,确定最佳酶用量和酶解时间分别为300 U和36 h,利用玉米芯酶解液进行深层液体发酵,第20天虫草素产量达到(25.53±3.27)mg/L,表明玉米芯适用于虫草素发酵生产。The effects of glucose and xylose as carbon sources on cordycepin production were investigated during submerged liquid fermentation with C.militaris,and the feasibility of corncob as carbon source for cordycepin production was studied.The results showed that when glucose or xylose was the only carbon source of Cordyceps militaris,the optimum carbon source were glucose 30 g/L or xylose 20 g/L,and the yield of cordycepin reached(144.63±19.48)mg/L and(50.61±0.66)mg/L,respectively.Two-stage fermentation of glucose xylose was established.The production intensity of cordycepin reached(3.55±0.09)mg/(L·d),40.32%higher than that of 20 g/L xylose fermentation system.The enzymatic hydrolysis conditions of corncob were optimized as the enzyme dosage of 300 U and reaction time of 36 h,respectively.The submerged liquid fermentation was carried out using the enzymatic hydrolysate of corncob,where cordycepin titer reached(25.53±3.27)mg/L after 20 d,indicating that corncob is suitable for cordycepin fermentation production.
