摘  要:本研究以甜玉米为材料,分别对化学、超声波、微波、酶法4种花青素提取方法进行优化,综合比较发现,微波提取方法的花青素吸光度值最大(1.072),花青素含量达到560 mg/kg,提取效果最佳,酶法辅助提取的方法不佳。对黑甜玉米杂交组合BS634×SP5029和W55×SD-4鲜子粒花青素组分分析发现,BS634×SP5029含有天竺葵素3-O-葡萄糖苷(41.15%)、芍药花素3-O-葡萄糖苷(29.70%)、矢车菊素3-O-半乳糖苷(3.77%)等17种已知花青素代谢物;W55×SD-4含有芍药花素3-O-葡萄糖苷(33.02%)、天竺葵素3-O-葡萄糖苷(29.41%)、矢车菊素O-丁香酸(8.12%)等17种已知花青素代谢物。两个黑甜玉米自交系SP5029和SD-4组配的杂交组合,参加四川省甜玉米联试,表现优质、抗病,鲜子粒花青素含量分别高达675 mg/kg和719 mg/kg,2020年通过四川省农作物品种审定,定名为黑甜玉8号(BS634×SP5029)和黑甜玉13号(W55×SD-4)。In this study,using sweet corn as material,four extraction methods of anthocyanin,chemical extraction,ultrasonic extraction,microwave extraction and enzymatic extraction,were compared as well as optimizing four methods respectively.The comprehensive comparison showed that the microwave extraction method had the highest absorbance value(1.072),and the content of anthocyanin reached to 560 mg/kg,which has the highest extracting effect among four methods,while the enzymatic extraction method has worse extracting effect.Based on the analysis of anthocyanin components in fresh kernels of BS634×SP5029 and W55×SD-4,two black sweet corn combinations.It was found that BS634×SP5029 contained 17 different anthocyanin components,including geranium 3-o-glucoside(41.15%),paeoniflorin 3-o-glucoside(29.70%)and cornvalin 3-o-galactoside(3.77%).W55×SD-4 contains 17 known anthocyanin metabolites,such as paeoniflorin 3-o-glucoside(33.02%),geranium 3-o-glucoside(29.41%),and cornflower o-eugenic acid(8.12%).In addition,two black sweet corn combinations,BS634×SP5029 and W55×SD-4,crossed by two black sweet corn inbred lines SP5029 and SD-4 used by this study,passed through Sichuan province variety trial for fresh corn.They perform high quality,resistance disease,and high anthocyanin contents(675 mg/kg and 719 mg/kg)in fresh kernels,both of which was released as Black Sweet Corn No.8(BS634×SP5029)and Black Sweet Corn No.13(W55×SD-4)in 2020.
