摘  要:目的:建立一种高效液相色谱法测定保健食品胶囊壳中7种合成着色剂含量的方法。方法:用60 ℃热水溶解胶囊壳,通过聚酰胺SPE固相萃取柱富集洗脱,得到含有合成着色剂的待测样,经安捷伦ZORBAX C;色谱柱分离,甲醇-乙酸铵溶液作为流动相梯度洗脱,各合成着色剂的特征波长为检测波长,DAD检测器检测,外标法定量。结果:7种合成着色剂分离效果良好,在1~20 μg/mL呈现良好线性关系,相对系数均大于0.999 0,回收率为86.42%~96.70%,RSD均小于1%。检出限为0.5 mg/kg,定量限为2.0 mg/kg。采用该方法对13批明胶空心胶囊和4批保健食品胶囊壳进行测定,均检测出国标允许使用的合成着色剂。结论:该方法操作简便、准确可靠,适用于保健食品胶囊壳中合成着色剂的测定。Objective:To establish a method for the determination of seven synthetic colorants in health food capsule shell by HPLC.Methods:the capsule shell was dissolved in 60 ℃ hot water,enriched and eluted by polyamide SPE solid phase extraction column to obtain the sample to be tested containing synthetic colorant.It was separated by Agilent ZORBAX C;chromatographic column,and gradient eluted by methanol ammonium acetate solution as mobile phase.The characteristic wavelength of each synthetic colorant was the detection wavelength,detected by DAD detector and quantified by appearance method.Results:the separation effect of the seven synthetic colorants is good,showing a good linear relationship in the range of 1~20 μ g/mL,the relative coefficients are greater than 0.999 0,the recovery is 86.42%~96.70%,and the RSD is less than 1%.The detection limit is 0.5 mg/kg and the quantitative limit is 2.0 mg/kg.13 batches of gelatin hollow capsules and 4 batches of health food capsule shells were determined by this method,and the synthetic colorants allowed by the national standard were detected.Conclusion:the method is simple,accurate and reliable.It is suitable for the determination of synthetic colorants in health food capsule shell.
