摘 要:对芦柑叶多酚的超声波辅助提取工艺进行响应面法优化,并研究芦柑叶多酚对亚硝酸盐的清除活性。先考察了乙醇浓度、超声时间、液料比和超声温度四个单因素对芦柑叶多酚提取率的影响,再结合响应面法对提取工艺条件进行优化,最后考察了芦柑叶多酚对亚硝酸盐的清除能力。结果表明,芦柑叶多酚的最佳提取工艺为:乙醇浓度61%,超声时间50 min,液料比20 mL/g和超声温度63℃,测得芦柑叶多酚的提取率为32.27 mg/g,其与模型预测的理论值(32.58 mg/g)相对误差为0.95%,说明所建立的模型方程准确、可靠,可应用于芦柑叶多酚提取工艺的预测。在所考察的质量浓度范围内,芦柑叶多酚均能较好地清除亚硝酸盐。多酚的质量浓度与亚硝酸盐的清除率成正比,芦柑叶多酚对亚硝酸盐清除的IC_(50)为1.93 mg/L,为芦柑叶资源开发成一种新型的天然亚硝酸盐清除剂提供了科学依据。The ultrasonic-assisted extraction process of polyphenols from Ponkan leaves was optimized by response surface methodology,and the scavenging activity of polyphenols on nitrite was studied.Firstly,the effects of four single factors of ethanol concentration,ultrasonic time,liquid-to-material ratio and ultrasonic temperature on the extraction yield of polyphenols from Ponkan leaves were investigated.Then,the extraction process conditions were optimized with response surface methodology.Finally,the ability of Ponkan leaves polyphenols to remove nitrite was investigated.The results showed that the optimal extraction process for the polyphenols from Ponkan leaves was as follows:ethanol concentration was 61%,ultrasonic time was 50 min,liquid-to-material ratio was 20 mL/g,and ultrasonic temperature was 63℃.The extraction yield of polyphenols from Ponkan leaves was 32.27 mg/g,its relative error with the theoretical value 32.58 mg/g predicted by the model was 0.95%,indicating that the established model equation was accurate and reliable,and could be applied to the prediction of the extraction process of Ponkan leaves polyphenols.Within the concentration range investigated,the polyphenols in the Ponkan leaves could remove nitrite well,and the concentration of polyphenols was directly proportional to the removal rate of nitrite.The IC_(50) of Ponkan leaves polyphenols for nitrite removal was 1.93 mg/L,which provided a scientific basis for the development of Ponkan leaves resources into a new type of natural nitrite scavenger.
