正交法优化大孔树脂提纯菝葜中绿原酸的工艺

摘  要:试验旨在研究大孔树脂提纯菝葜中绿原酸的工艺。以菝葜为原料,利用超声波粗提,经大孔树脂D101除杂,通过正交试验的方法研究大孔树脂提纯菝葜中绿原酸的静态吸附和解吸工艺,结合动态试验,得出大孔树脂提纯绿原酸的最优工艺:大孔树脂D101静态除杂,用大孔树脂NKA-Ⅱ吸附,在pH 1、料液浓度0.08 mg/mL、上样液流速2 BV/h条件下,上样8 BV达绿原酸泄漏点,用pH 3的酸水2 BV冲洗后,用40%乙醇溶液解吸,收集第3~第5 BV解吸液,解吸流速为2 BV/h。在此工艺条件下,菝葜绿原酸含量由粗提液中的0.412 2%提高到25.06%,产率为0.035%。大孔树脂NKA-Ⅱ再生处理后,共计可使用8次。该试验可为菝葜中绿原酸的工业生产提供数据参考。The purification of chlorogenic acid from Smilax china by macroporous resin was studied.Taking Smilax china as raw material,the method of ultrasonic wave was used to extract chlorogenic acid from Smilax china,and the macroporous resin D101 was used to remove impurities.The static adsorption and desorption process of chlorogenic acid from Smilax China were studied by orthogonal test.Combined with dynamic test,the optimal process of purification chlorogenic acid from Smilax china by macroporous resin D101 was obtained as follows:static impurity removal by macroporous resin D101,adsorption by macroporous resin NKA-Ⅱ,pH 1,material concentration 0.08 mg/mL,the flow rate of sample solution 2 BV/h,8 BV of sample solution reached the leakage point of chlorogenic acid.After washing with pH 3 acid water 2 BV,desorbing with 40% ethanol solution,and collecting the desorption solution of the 3^(rd)-5^(th) BV.The desorption flow rate was 2 BV/h.Under this condition,the content of chlorogenic acid in Smilax china increased from 0.412 2% to 25.06%,and the yield was 0.035%.After regeneration,NKA-Ⅱ could be used for 8 times.The data reference for the industrial production of chlorogenic acid in Smilax china could be provided.
