摘 要:针对目前赭曲霉毒素A(OTA)的检测方法流程烦琐、耗时长等问题,提出一种基于弱磁检测和免疫技术结合的新型OTA定量检测方法。通过在纳米磁珠(MNPs)表面修饰OTA抗体,获得具有免疫活性的磁珠抗体复合物,在亥姆霍兹线圈产生的匀强磁场下,使用磁力计对磁性层析试纸条检测线(T线)和质控线(C线)上MNPs的弱磁信号进行检测,提取T线和C线上响应磁信号的比值(T/C),分析T/C随OTA质量浓度变化的趋势并获取标准化曲线。试验结果表明:MNPs响应信号检测结果同线磁偶极子模型仿真结果基本一致,OTA质量浓度的对数与磁信号之间呈现很好的线性相关性,试纸条的定性检测限为0.10 ng/mL,定量检测限为0.32 ng/mL,线性检测范围为0.32~5 ng/mL,完成1次检测的时间仅需30 s。该检测方法可用于现场即时检测,具有很好的应用前景。In view of the present ochratoxin A(OTA)method in the problems of complex and time-consuming process,a new quantitative OTA detection method based on the combination of weak magnetic signal detection technology and immunoassay technology was proposed.By modifying the surface of magnetic nanoparticles(MNPs)with OTA antibodies,a magnetic bead antibody complex with immune activity can be obtained.Under a magnetic field of uniform intensity generated by the Helmholtz coil,a magnetometer was used to detect the weak magnetic signal generated by the MNPs on the test line(T line)and the quality control line(C line)on the magnetic chromatography papers,and the ratio of the T∶C line response magnetic signal,namely T/C value,was extracted.Through analyzing the T/C values with OTA concentration,a standardization curve can be obtained.The results showed that the detection results of MNPs response signal were basically consistent with the simulation results of linear magnetic dipole model,and a good linear correlation between OTA mass concentration and magnetic signal was obtained.The limit of the qualitative detection of the test strip was 0.10 ng/mL,the quantitative detection limit was 0.32 ng/mL,and the linear detection was 0.32-5 ng/mL.Thirty seconds were enough for completing a single test.This detection method can be used for real-time detection in the field and has a good application prospect.
