摘 要:为获得高效园林废弃物降解菌,采用沙氏培养基和LB培养基进行真菌和细菌富集,利用CMC-Na水解圈测定法初筛,利用DNS法测定初筛菌株的内切型-β-葡聚糖酶活(CMC酶活)和外切型-β-葡聚糖酶活,最后对菌株进行分子生物学鉴定。结果表明:筛选得到6株酶活较高的园林废弃物降解细菌,CMC酶活为22.5~34.5 U/mL;外切型-β-葡聚糖酶活为2.9~6.2 U/mL。其中:CMC酶活最高的是14号菌株,为芽孢杆菌属(Bacillus sp.);外切型-β-葡聚糖酶活最高的是28号菌株。成功鉴定4株菌株的种属,分别为14号和29号为芽孢杆菌属(Bacillus spp.)、20号为假单胞菌属(Pseudomonas sp.)、30号为泛菌属(Pantoea sp.)。本研究通过筛选、鉴定获得4株高效园林废弃物降解菌株,在此基础上与同类降解菌进行比较分析,为进一步开展高效园林废弃物降解菌种制备提供了基础和技术支持。In order to obtain high-efficiency garden waste degrading bacteria, this paper used Sabouraud medium and LB medium to enrich fungi and bacteria, used the CMC-Na hydrolysis circle assay method for preliminary screening,and adopted the DNS method to determine endo-β-glucanase activity(CMC activity) and the exo-β-glucanase activity of each strain. Finally, the strains were identified by molecular biology. The results showed that six strains of garden waste degrading bacteria with high enzyme activity were screened, their CMC activities were 22.5-34.5 U/mL;the exo-β-glucanase activities were 2.9-6.2 U/mL. The CMC activity was the highest in No. 14 strain, which belonged to Bacillus sp., and the exo-β-glucanase activity was the highest in No. 28 strain. Four strains were identified successfully, No. 14 and No. 29 were Bacillus spp., No. 20 was Pseudomonas sp., and No. 30 was Pantoea sp. Through screening and identification, four high-efficiency garden waste degrading strains were obtained. On this basis, comparison and analysis with similar degrading bacteria were carried out to provide basic and technical support for the further development of the preparation of high-efficiency garden waste degrading strains.
