基于GEO数据库的肾上腺皮质腺癌关键基因筛选及预后分析

摘  要:目的探究与肾上腺皮质腺癌发生发展相关的关键基因并进行预后分析,挖掘潜在诊断和治疗靶点。方法利用生物信息学技术对GSE12368、GSE19750、GSE143383、GSE90713、GSE14922、GSE75415共6个数据集行差异基因表达分析,获取共同差异表达基因,STRING数据库针对差异基因行蛋白表达网络分析,并运用Cytoscape软件cytoHubba插件获取排名前5的5个关键基因。对5个关键基因行GO、KEGG富集分析,并利用GEPIA2工具行生存分析。结果6个数据集差异基因分析取交集得到差异基因共16个,其中上调基因5个,下调基因11个,cytoHubba插件共筛选出5个关键基因(CCNB1、TOP2A、TPX2、ZWINT、NCAPG),均为上调基因。富集分析提示5个关键基因参与细胞周期检查点、染色体分离、DNA构象改变、有丝分裂细胞周期负调控、细胞器合成等多种过程,且生存分析提示5个关键基因在肾上腺皮质腺癌中高表达,与较差的预后相关。结论CCNB1、TOP2A、TPX2、ZWINT、NCAPG与肾上腺皮质腺癌预后密切相关,是其潜在的诊断标志物及治疗靶点。Objective To identify the hub genes related to the occurrence and development of adrenal cortical adenocarcinoma(ACC),and analyze the prognosis of ACC.Methods The differentially expressed genes(DEGs)were screened based on the gene chip datasets GSE12368,GSE19750,GSE143383,GSE90713,GSE14922 and GSE75415.Protein-protein interaction(PPI)network was analyzed with the STRING database,Cytoscape and cytoHubba software,and the top 5 hub genes were obtained.GO analysis and KEGG enrichment pathway analysis were performed.The prognostic analysis of the 5 hub genes was analyzed with GEPIA2.Results Altogether 16 DEGs were obtained from the 6 datasets,of which 5 were up-regulated and 11 were down-regulated.The top 5 were CCNB1,TOP2A,TPX2,ZWINT and NCAPG.Enrichment analysis found that the 5 DEGs were related to cell cycle checkpoint,chromosome segregation,DNA conformation change,negative regulation of mitotic cell cycle and organelle fission.They were highly expressed in ACC and indicated poor prognosis.Conclusion CCNB1,TOP2A,TPX2,ZWINT and NCAPG are related to the prognosis of ACC and may become potential diagnostic markers and therapeutic targets.
