摘 要:为确定蛹虫草产虫草素放大工艺条件,考察了菌种培养质量和发酵高径比对虫草素合成的影响。确定种子培养最佳条件为:转速250 r/min,接种量15%,扩培级数2次,此条件下虫草素发酵产量可达6 g/L左右。另外,装液量最适高径比为2 cm/30 cm。在此条件下,进行了120 L多层反应器发酵工艺验证。发酵25 d,虫草素产量为5.32 g/L,转化率57%,与小试工艺结果基本一致。实验结果表明,工艺放大具有应用可行性,为进一步工业化生产提供了参考。In order to determine the amplification technology for cordycepin, the effects of cultivation quality and height-diameter ratio of fermentation on the synthesis of cordycepin were investigated. The results showed that the concentration of cordycepin reached 6 g/L when the optimal conditions of seed cultivation were as follows: the rotation speed of 250 r/min, the inoculum size of 15%, and the subculture times of two. In addition, the optimal height-diameter ratio of liquid loading was 2 cm/30 cm. Under these conditions, a 120 L multi-layer reactor was used to verify the fermentation process. After 25 days of fermentation, the concentration of cordycepin was 5.32 g/L and the conversion rate was 57%, which was consistent with the results of small scale test. The experimental results showed that the process scaling up is feasible, which provides a reference for further industrialization.
