摘 要:γ-聚谷氨酸(γ-Polyglutamic acid,简称γ-PGA)是一种具有生物相容性、生物可降解性和无毒无污染的高分子聚合物。微生物发酵法生产γ-PGA工艺简单、产量较高,是γ-PGA的主要生产方法,但是γ-PGA发酵液成分复杂、黏度高,从发酵液中分离纯化γ-PGA存在一定难度。该工艺以枯草芽孢杆菌为发酵菌株,以谷氨酸钠为原料进行发酵生产γ-聚谷氨酸、优化发酵及提取工艺。实验结果表明:当发酵温度为32℃时,γ-PGA产量较高;当发酵液中谷氨酸钠质量分数为4.2%时,发酵液黏度可达到最高;采用乙醇入料提纯方式可有效去除发酵液中的色素、大分子蛋白和其他有机杂质。Gama-Polyglutamic acid(γ-PGA)is a kind of high molecular polymer with biocompatible,biodegradable,non-toxic,and pollution-free.The microbial fermentation method becomes the main way forγ-PGA production owing to its simple and high yield.However,due to the complex composition and high viscosity ofγ-PGA fermentation broth,separation and purification ofγ-PGA from the fermentation broth is difficult.In this study,γ-PGA was produced by Bacillus subtilis with glutamate sodium as raw material.The fermentation and extraction processes were optimized.The results showed that the yield ofγ-PGA was higher when the fermentation temperature was 32℃.The viscosity of fermentation broth reached the highest when glutamate content was 4.2%.Adding ethanol to purify fermentation broth can effectively remove pigments,macromolecular proteins,and other organic impurities.
