摘 要:为选择鲜乳和复原乳鉴别方法,采用双向电泳指纹图谱分析技术分析鲜乳和复原乳总蛋白。通过不同胶条比对分析发现,pH4.7~5.9的17 cm胶条等电聚焦后能够有效区分蛋白点,最适合牛乳蛋白电泳;选用pH4.7~5.9的17 cm胶条,对市场上购买的巴氏乳以及巴氏处理的复原乳进行电泳分析,发现电泳图谱与自制参照乳图谱基本一致。结果表明,该方法是一种行之有效的区分鲜乳和复原乳的检测方法,具有分辨能力强、重现性好等优点。In order to select an appropriate method to identify fresh milk from reconstituted milk,the total protein of the two milk was analyzed by the two-dimensional electrophoresis fngerprint analysis.It was found that,through comparison and analysis of diferent adhesive strips,protein spots could be distinguished by 17 cm strip with pH4.7-5.9 when it was isoelectric focusing,which was most suitable for milk protein electrophoresis;the pasteurized milk purchased from markets and the pasteurized reconstituted milk were analyzed by electrophoresis by using 17 cm strip with pH4.7-5.9,it was found that the electrophoretogram was basically consistent with the self-made reference milk map.In conclusion,the method was efective to distinguish fresh milk from reconstituted milk due to its advantages such as high resolution and good reproducibility.
