摘 要:先天性心血管疾病是一类由于遗传、环境及妊娠期感染等多因素相互作用而导致的先天性疾病,但确切的病因迄今尚不清楚。随着研究的逐步深入,越来越多的证据表明遗传因素在其发病机制中发挥重要作用。在心脏胚胎发育的过程中涉及许多基因的参与,这些基因如果发生突变往往会导致先天性心血管疾病。同源建模作为一种通过计算机算法预测由于基因突变导致所形成的蛋白质结构及功能发生改变的技术,在先天性疾病发病机制研究中的应用已成为未来的趋势。该文旨在对同源建模技术在先天性心血管疾病发病机制研究中的应用进行综述。Congenital cardiovascular disease is a kind of congenital disease caused by the interaction of multiple factors such as heredity,environment and pregnancy infection,but its exact etiology is still unclear.With the deepening of research,more and more evidences show that genetic factors play a significant role in its pathogenesis.Many genes are involved in the development of heart embryos,and the mutation of these genes often leads to congenital cardiovascular diseases.Homology modeling is a technique to predict the structural and functional changes of proteins caused by gene mutation through computer algorithm,and its application in the pathogenesis of congenital diseases has become a future trend.The purpose of this paper is to review homology modeling and its application in pathogenesis of congenital cardiovascular diseases briefly.
