摘  要:目的探讨卵巢癌组织中SPOCK2基因的表达及其临床意义。方法使用基因表达谱动态分析(GEPIA)在线工具分析卵巢癌组织中SPOCK2 mRNA的表达情况,使用人类蛋白免疫组化表达数据库(The Human Protein Atlas)分析卵巢癌组织中SPOCK2蛋白的表达情况,通过UALCAN在线工具分析SPOCK2 mRNA在不同分化程度卵巢癌组织中的表达。使用GEPIA在线工具分析卵巢癌患者SPOCK2 mRNA表达水平及最佳截断值,依据最佳截断值将样本分为高表达组和低表达组并绘制生存曲线。使用TIMER在线工具分析卵巢癌浸润细胞类型及浸润程度,分析SPOCK2 mRNA表达与卵巢癌浸润细胞类型及浸润程度的相关性。使用SurvivalMeth在线工具分析SPOCK2基因甲基化水平,探讨其与卵巢癌预后的相关性。结果使用GEPIA及The Human Protein Atlas在线工具分析显示,与正常组织或癌旁组织比较,卵巢癌组织高表达SPOCK2 mRNA和SPOCK2蛋白(均P<0.05)。不同分化程度卵巢癌组织中SPOCK2 mRNA表达比较显示,SPOCK2 mRNA表达水平越高,卵巢癌分化程度越低。生存曲线分析显示,SPOCK2 mRNA表达与生存率显著相关,SPOCK2 mRNA高表达组总体生存期明显低于低表达组(P<0.01);SPOCK2 mRNA高表达组患者5年生存率仅为22%,低表达组5年生存率为39%。SPOCK2 mRNA表达水平与Th1细胞、Th2细胞、CD4+记忆性T细胞、M1型巨噬细胞浸润程度均呈负相关(r=-0.135、-0.214、-0.205、-0.147,均P<0.05),SPOCK2 mRNA表达水平与肿瘤相关巨噬细胞浸润程度呈正相关(r=0.289,P<0.01)。两种DNA甲基化探针检测均显示卵巢癌组织中SPOCK2基因甲基化水平较正常组织降低(P<0.01)。生存分析显示,高风险组和低风险组生存率具有显著差异,且高风险组卵巢癌组织中SPOCK2甲基化水平较低(均P<0.05)。结论SPOCK2在卵巢癌组织中高表达并促进疾病进展,是卵巢癌潜在的预后指标及治疗靶点。Objective To investigate the expression of SPOCK2 gene in ovarian cancer and its clinical significance.Methods The online software Gene Expression Profiling Interactive Analysis(GEPIA)was used to analyze the expression of SPOCK2 mRNA in ovarian cancer tissues.The online software Human Protein Atlas was used to analyze the expression of SPOCK2 protein in ovarian cancer tissues,and the online software UALCAN was used to analyze the expression of SPOCK2 mRNA in ovarian cancer tissues with different degrees of differentiation.The online software GEPIA was used to determine the optimal cut-off value of SPOCK2 mRNA expression in patients with ovarian cancer.According to the optimal cut-off value,the samples from these patients were divided into the high expression group and low expression group,with generation of a survival curve.The online software TIMER was used to examine the type of ovarian cancer infiltrating cells and the extent of infiltration.The correlation between SPOCK2 mRNA expression,the type of ovarian cancer infiltrating cells and the extent of infiltration was determined.The online software SurvivalMeth was used to analyze the methylation of SPOCK2 and its correlation with the prognosis of ovarian cancer.Results The online softwares GEPIA and The Human Protein Atlas,showed that,compared with normal or adjacent tissues,SPOCK2 mRNA and protein were highly expressed in ovarian cancer tissues(both P<0.05).In ovarian cancer tissues with different degrees of differentiation,higher level of SPOCK2 mRNA expression was associated with lower differentiation degree of ovarian cancer cells.Survival curve analysis showed that SPOCK2 mRNA expression was significantly correlated with survival rate,and the overall survival time was significantly lower in SPOCK2 mRNA high expression group than that in low expression group(P<0.01).The 5-year survival rate of patients in the SPOCK2 mRNA high expression group was only 22%,compared with 39%in the low expression group was 39%.The SPOCK2 mRNA expression level was negatively c
