摘 要:目的:通过网络药理学方法探讨固肾安胎方治疗先兆性流产的作用机制。方法:运用TCMSP、SwissTarget等数据库筛选化学成分、获取潜在靶点,构建药材-成分-靶点网络图,并进行信号通路的富集与分析,利用分子对接技术探究潜在靶点与活性成分的相互作用。结果:获得固肾安胎方的核心成分89个和作用靶点34个,涉及Steroidogenesis、PI3K-Akt等23条信号通路;分子对接结果表明槲皮素、山姜素、山柰酚与CYP1B1、ACHE、PGF等靶蛋白具有良好的结合能力。结论:网络药理学揭示了固肾安胎方可通过多个靶点、多个信号通路来治疗先兆性流产,为进一步研究其作用机制提供了理论基础。OBJECTIVE To explore the molecular mechanism of Gushen Antai formula in treatment of threatened abortion by network pharmacology.METHODS The active constituents and target proteins were obtained from TCMSP,SwissTarget databases,medicinal material-component-target network map was constructed,and the pathway enrichment was performed for molecular mechanism analysis,then,molecular docking was used to explore the interaction between potential targets and active components.RESULTS 89 active components with 34 potential targets were screened out,which were involved in 23 signal pathways,such as steroidogenesis,PI3K-Akt signal pathway.The docking results showed that main active ingredients of Gushen Antai formula had good binding abilities with key target proteins.CONCLUSION Through the network pharmacology,active components and the mechanism of Gushen Antai formula in the treatment of threatened abortion were revealed,which provided a theoretical basis for further research on its mechanism of action.
