摘 要:目的分析Hedgehog(Hh)通路的关键基因CDO兄弟同源物(BOC)、SMO、CDON、PTCH1、GAS1、HHIP、脑胶质瘤相关癌基因同源物(GLI)、SHH在脑胶质瘤中表达模式的相关性。方法按照常春藤胶质瘤数据库提供的数据指标及相关下载流程,结合脑胶质瘤组织学结构、临床参数和基因组数据等变量对目的基因的表达数据进行收集整理。对Hh通路的关键基因BOC、SMO、CDON、PTCH1、GAS1、HHIP、GLI、SHH在脑胶质瘤的不同病理分区、分子亚型、甲基化与否等条件下呈现的表达模式进行分析。所有数值均被数据和TbT归一化来处理,采用R编程语言绘制箱形图和热图,并进行统计分析。结果BOC与SMO在脑胶质瘤的不同病理分区、分子亚型及异柠檬酸脱氢酶(IDH)突变的条件下表达模式高度一致;SHH与GLI在肿瘤组织的不同病理分区及分子亚型中表达差异无统计学意义(P>0.05);CDON与PTCH1在脑胶质瘤不同分子亚型中表达模式基本一致,且IDH基因突变组CDON与PTCH1的表达水平显著高于IDH基因野生型组(P<0.05)。O_(6)-甲基鸟嘌呤-DNA甲基转移酶(MGMT)基因甲基化组的GAS1、BOC、SHH表达水平高于MGMT基因非甲基化组(P<0.05);PTCH1、HHIP、GLI表达水平低于MGMT基因非甲基化组(P<0.05)。结论Hh通路上8个关键基因在脑胶质瘤的不同病理分区、分子亚型、IDH突变及MGMT甲基化与否条件下的表达模式存在明显差异,依据上述基因的表达变化结合临床因素能够更全面地评估脑胶质瘤患者的预后。Objective To analyze the correlation between the expression patterns of eight key Hedgehog(Hh)signaling genes of brother of CDO(BOC),Smoothened(SMO),cell adhesion associated,oncogene regulated(CDON),Patched 1(PTCH1),growth arrest specific 1(GAS1),Hedgehog interacting protein(HHIP),glioma-associated oncogene homolog(GLI),Sonic Hedgehog(SHH)in glioma.Methods According to the data indicators and related download process provided in the official webpage of the Ivy Glioblastoma Atlas Project dataset,the expression data of target genes were collected and arranged in combination with the variables such as glioma histological structure,clinical parameters and genome data.The expression patterns of key Hh signaling genes such as BOC,SMO,CDON,PTCH1,GAS1,HHIP,GLI and SHH were analyzed under the conditions of different pathological zoning,glioma molecular subtypes,methylation or not.All the values were processed by data and TbT normalization.The R programming language was used to draw box diagrams and heat maps,and statistical analysis was performed.Results The expression patterns of BOC and SMO were highly consistent under different pathological subtypes,molecular subtypes and isocitrate dehydrogenase(IDH)gene mutation in glioma.There was no significant difference in the expression of SHH and GLI in different pathological divisions and molecular subtypes of tumor tissues(P>0.05).The expression patterns of CDON and PTCH1 in different subtypes of glioma were basically consistent,and the expression of CDON and PTCH1 in IDH gene mutation group was significantly higher than that in IDH gene wild-type group(P<0.05).The expression levels of GAS1,BOC and SHH in O_(6)-methylguanine-DNA methyl-transferase(MGMT)gene methylated group were higher than those in MGMT gene non-methylated group(P<0.05);the expression levels of PTCH1,HHIP and GLI were lower than those in MGMT gene non-methylated group(P<0.05).Conclusion The expression patterns of eight key genes in the Hh pathway are significantly different in glioma under different conditio
