摘 要:目的探究APP(淀粉样前体蛋白)的表达在TGFβ诱导的前列腺癌PC-3细胞的迁移和侵袭中的作用。方法常规培养前列腺癌PC-3细胞后,使用浓度为20 ng/ml的TGFβ处理细胞48 h。通过Transwell迁移小室观察TGFβ对前列腺癌PC-3细胞迁移和侵袭能力的影响。通过蛋白印迹和qPCR方法检测TGFβ处理后细胞APP的表达水平。通过蛋白印迹实验检测TGFβ下游分子Smad 1和Smad 2的活化水平,检测信号通路相关蛋白的表达水平。使用smad信号通路抑制剂LY2109761作用后,检测APP的表达水平,同时检测PC-3细胞的迁移和侵袭能力。结果20 ng/ml的TGFβ处理能够显著提升前列腺癌PC-3细胞的迁移和侵袭能力。蛋白印迹和qPCR方法都发现TGFβ能够显著提升PC-3细胞APP的表达水平(P<0.05)。在此过程中,TGFβ下游分子Smad 1和Smad 2的表达显著被上调(P<0.05),其下游信号分子活化水平同样显著上调(P<0.05)。使用smad信号通路抑制剂LY2109761作用后,APP的表达水平显著降低,同时PC-3细胞的迁移和侵袭能力也显著降低。结论TGFβ显著提升前列腺癌PC-3细胞的迁移和侵袭能力,该效应可能通过促进APP的表达产生。Objective To explore impact of APP on the TGFβ-induced prostate cancer PC-3 cells'migration and invasion.Methods After routine culture of prostate cancer PC-3 cells,cells were treated with TGFβat a concentration of 20 ng/ml for 48 h.The effect of TGFβon the migration and invasion of prostate cancer PC-3 cells was observed by Transwell chamber.The expression level of APP after TGFβtreatment was detected by Western blot and qPCR.The activated levels of Smad 1 and Smad 2 of TGFβdownstream molecular were detected by Western blotting,and the expression levels of signal pathway-related proteins were detected.After treated with smad signaling pathway inhibitor LY2109761,the expression level of APP was detected,and the migration and invasion ability of PC-3 cells were detected.Results TGFβtreatment at 20 ng/ml significantly increased the migration and invasion of prostate cancer PC-3 cells.Both Western blot and qPCR methods found that TGFβsignificantly increased APP expression in PC-3 cells(P<0.05).During this process,the expression of Smad 1 and Smad 2 downstream molecules of TGFβwas significantly up-regulated(P<0.05).The activation level of downstream signaling molecules was also significantly up-regulated(P<0.05).After the action of smad signaling pathway inhibitor LY2109761,the expression level of APP was significantly decreased,and the migration and invasion ability of PC-3 cells were also significantly reduced.Conclusion TGFβsignificantly enhances the migration and invasion of prostate cancer PC-3 cells,which may be caused by the promotion of APP expression.
