升清降浊胶囊对db/db小鼠肾组织PDGF-B表达的影响

摘　　要：目的:观察升清降浊胶囊改善自发性2型糖尿病db/db小鼠肾损伤的作用及对肾组织血小板衍化生长因子-B(PDGF-B)表达的影响。方法:将16只8周龄雄性db/db小鼠随机分为db/db模型组(n=8)和升清降浊组(n=8),选取db/m小鼠为空白对照组(n=8),升清降浊组给予升清降浊胶囊1800 mg·kg^(-1)·d^(-1)灌胃,db/db模型组和db/m组小鼠以等量生理盐水灌胃,干预8周后采集各组小鼠血液、尿液检测空腹血糖、血肌酐和尿蛋白定量(ACR)等,处死各组小鼠,HE染色、PAS染色观察肾脏组织病理形态变化,免疫组化法检测小鼠肾PDGF-B和PDGFR-β表达。结果:升清降浊组小鼠尿蛋白定量、血糖、血肌酐水平明显降低(P<0.05),肾脏病理结果显示系膜及基质增生情况明显减轻;肾组织PDGF-B和PDGFR-β表达低于db/db模型组,但尚未达到db/m组水平。结论:升清降浊胶囊可明显改善db/db小鼠肾损伤,延缓糖尿病肾病进展,减轻肾组织PDGF-B和PDGFR-β表达。Objective:To observe the effects of Shengqing Jiangzhuo capsule on renal injury and expression of platelet-derived growth factor-B(PDGF-B)in renal tissue of db/db mice model of spontaneous type 2 diabetes.Methods:Sixteen male db/db mice aged 8 weeks old were randomly divided into two groups including model group(n=8)and Shengqing Jiangzhuo group(n=8).Another eight db/m mice were used as blank control group(n=8).Shengqing Jiangzhuo group were given Shengqing Jiangzhuo capsule 1800 mg·kg^(-1)·d^(-1),model group and db/m group were given the same amount of normal saline.Continuous intervention for 8 weeks,the fasting blood glucose(FBG),serum creatinine(Scr)and urinary microalbumin/urine creatinine ratio(ACR)level of each group were detected.The mice in each group were killed,the kidneys were separated to observe the pathomorphological changes with hematoxylin-eosin and periodic acid Schiff staining,and detect the expression of PDGF-B and PDGFR-βwith immunohistochemistry.Results:Compared with the model group,the levels of ACR、FBG and Scr were significantly reduced in Shengqing Jiangzhuo group(P<0.05),and the proliferation of Mesangial and Matrix in renal pathology was significantly reduced in Shengqing Jiangzhuo group.The expression of PDGF-B and PDGFR-βin renal tissue was lower than model group,but higher than the db/m group.Conclusion:Shengqing Jiangzhuo capsule can obviously reduce the renal damage of db/db mice and delay the progression of diabetic nephropathy,The mechanism may be related to the reduction of PDGF-B and PDGFR-βexpressions in renal tissues.
