摘　　要：目的:基于生物信息学挖掘阐明ERBB3在结肠腺癌中的表达及临床意义。方法:检索Oncomine、TCGA、String-DB数据库等基因数据库,分析ERBB3在正常组织与结肠腺癌的表达差异,用R-sdutio进行患者生存及复发分析,并对与ERBB3相关的分子进行分析和预测。结果:(1)对ERBB3在结肠腺癌中365项研究数据进行差异性分析发现,ERBB3在结肠腺癌中表达明显增高;(2)在结肠腺癌中ERBB3 mRNA上调比ERBB3 mRNA下调多;(3)ERBB3mRNA表达与DNA拷贝数改变状态呈显著正相关;(4)在ERBB3高表达组较低表达组结肠腺癌复发率高。结论:ERBB3可能成为结肠腺癌的诊断及减少复发的新靶点,对其分子机制的研究具有重要意义。Objective:To demonstrate the expression and clinical significance of ERBB3 in colon adenocarcinoma based on bioinformatics data mining.Methods:Search the Oncomine,TCGA,String-DB database and other gene databases to analyze the expression difference of ERBB3 in normal tissues and colon adenocarcinoma.R-sdutio was used to analyze the survival and recurrence of patients,and the molecules related to ERBB3 were analyzed and predicted.Results:(1)Differential analysis of 365 data of ERBB3 in colon adenocarcinoma showed that the expression of ERBB3 was significantly increased in colon adenocarcinoma,compared with normal tissue.(2)The alteration frequency Up-regulation of ERBB3 mRNA is more than down-regulated in colon adenocarcinoma,rectal adenocarcinoma,and colorectal mucinous adenocarcinoma.(3)There was a significant positive correlation between ERBB3 mRNA expression and DNA copy number change status.(4)The high expression group of ERBB3 had higher recurrence rate,compared with the low expression group.Conclusion:ERBB3 may be a new target for diagnosis and recurrence reduction of colorectal adenocarcinoma,which is of great significance for the study of its molecular mechanism.
