摘  要:目的:分析长链非编码RNA(LncRNA)和程序性死亡配体1(PD-L1)表达与卵巢癌患者化疗耐药性的关系。方法:收集2019年1月-2020年12月于暨南大学附属第一医院就诊的128例卵巢癌患者临床资料进行回顾性分析,其中铂类药物耐药患者69例设为耐药组,铂类药物敏感患者59例设为敏感组,比较两组的卵巢癌组织中LncRNA与PD-L1的表达情况,分析患者LncRNA和PD-L1的表达与卵巢癌患者化疗耐药性的关系。结果:对LncRNA的两个分子[生长阻滞特异性转录因子5(GAS5)、HOXC基因座转录因子(HOTAIR)]进行分析,耐药组以上两个LncRNA分子的表达水平均明显高于敏感组(P<0.05),耐药组PD-L1表达水平明显高于敏感组(P<0.05);多因素logistic回归分析显示,GAS5、HOTAIR与PD-L1均为卵巢癌患者化疗耐药的独立危险因素(P<0.05);PD-L1表达水平和GAS5、HOTAIR表达水平均呈显著正相关(P<0.05);肿瘤细胞平均抑制率为(36.63±6.53)%,与PD-L1、GAS5、HOTAIR表达水平均呈负相关(P<0.05)。结论:卵巢癌组织中LncRNA的GAS5、HOTAIR分子与PD-L1的高表达与卵巢癌患者化疗耐药性密切相关,LncRNA与PD-L1相互协同,促进肿瘤细胞耐药。Objective:To analyze the relationship between the expression of long non-coding RNA (LncRNA) and programmed death ligand 1 (PD-L1) and chemoresistance in patients with ovarian cancer.Method:The clinical data of 128 patients with ovarian cancer treated in the First Affiliated Hospital of Ji’nan University from January 2019 to December 2020 were collected and analyzed retrospectively,among them,69 patients with platinum drug resistance were set as the drug-resistance group and 59 patients with platinum drug sensitivity were set as the sensitive group.The expression of LncRNA and PD-L1 in ovarian cancer tissues of the two groups were compared,the association between LncRNA and PD-L1 expression and chemoresistance in patients with ovarian cancer was analyzed.Result:Two molecules of LncRNA[growth arrest specific transcription factor 5 (GAS5) and HoxC locus transcription factor (HOTAIR)]were analyzed,the expression levels of these two LncRNA molecules in drug-resistant group were significantly higher than those in sensitive group (P<0.05),and the expression level of PD-L1 in drug-resistant group was significantly higher than that in sensitive group (P<0.05);multivariate logistic regression analysis showed that GAS5,HOTAIR and PD-L1 were independent risk factors of chemotherapy resistance in ovarian cancer patients (P<0.05).The expression level of PD-L1 was significantly positively correlated with the expression levels of GAS5 and HOTAIR (P<0.05);the average inhibition rate of tumor cells was (36.63±6.53)%,which was negatively correlated with the expression levels of PD-L1,GAS5 and HOTAIR (P<0.05).Conclusion:The high expression of GAS5,HOTAIR in LncRNA and PD-L1 of ovarian cancer is closely related to chemotherapy resistance of ovarian cancer patients,LncRNA and PD-L1 cooperate with each other to promote drug resistance of tumor cells.
