摘 要:建立以pH示差法检测苹果皮花色苷含量的方法,为苹果中花色苷的定量分析及苹果深加工提供基础数据,以60%乙醇溶液为提取剂,确定提取料液比、提取方式、提取温度、提取时间、氮吹浓缩条件;以pH 1.0和pH 4.5为缓冲体系,确定平衡温度和时间、最大吸收波长,在此条件下进行精密度试验。结果表明苹果皮花色苷提取料液比1∶10(g/mL),提取方式为400 W超声波辅助、温度40℃、时间50 min,提取两次并合并提取液,如需浓缩至适当浓度可在25℃下氮吹,反应平衡温度为40℃、平衡时间为70 min,最大吸收波长为505 nm。在此条件测得苹果皮花色苷含量为24.9 mg/100 g,RSD为4.5%。该方法准确可靠,可用于苹果皮花色苷含量的测定。A method for the determination of anthocyanin content in apple skin by pH differential method was established to provide basic data for quantitative analysis of anthocyanin and further processing of apple.60%ethanol solution was used as extractant to determine the solid-liquid ratio,extraction method,extraction temperature,extraction time and nitrogen blowing concentration conditions.The equilibrium temperature and time,maximum absorption wavelength were determined with pH 1.0 and 4.5 as buffer systems,and the precision test was carried out under this condition.Results show that the solid-liquid ratio of apple skin anthocyanin extraction was 1∶10(g/mL),the extraction method was 400 W ultrasonic assisted,the temperature was 40℃,the time was 50 min,extracted twice and combined.The extract could be nitrogen-blown at 25℃to concentration if necessary.The reaction equilibrium temperature was 40℃,the equilibrium time was 70 min,and the maximum absorption wavelength was 505 nm.Under these conditions,the anthocyanin content in apple skin was 24.9 mg/100 g,and the relative standard deviation was 4.5%.The method is accurate and reliable,and can be used for the determination of apple skin anthocyanin content.
