汉麻籽粕酶解物中抗肝癌Hep3B细胞活性肽的分离与鉴定

摘　　要：从汉麻籽粕的复合蛋白酶酶解产物中分离鉴定具有抑制肝癌Hep3B细胞活性的抗癌肽。用超滤系统对其酶解产物进行分离,研究多肽对肝癌Hep3B细胞的活性,采用MALDI-TOF/TOF质谱分析多肽超滤分离组分的分子质量分布范围,对高峰值信号峰进行结构表征,再通过合成肽对其结构与抗肝癌活性进行验证。获得2个抑制肝癌Hep3B细胞活性的抗肝癌肽,其氨基酸序列分别为YGRDEISVF和NKSGTYSNDDLSH。合成肽YGRDEISVF和NKSGTYSNDDLSH的分子质量分别为1 085.20、1 437.20 u,纯度分别为98.90%和98.63%,汉麻籽粕复合酶酶解产物含有抗肝癌肽类成分,为其抗肝癌肽的开发提供参考。Anticancer peptides inhibiting the activity of hepatocarcinoma Hep3 B cells were isolated and identified from the compound protease hydrolysate of hemp seed. Anticancer active peptides in the hydrolysates were purified by using the ultrafiltration membrane system to study the activity of hemp seed peptides on liver cancer Hep3 B cells. The molecular weight distribution range of the hemp seed peptide ultrafiltration fractions was analyzed by MALDI-TOF/TOF mass spectrometry. The structure of high peak signal peak was characterized, and its structure and anti-liver cancer activity were verified by synthetic peptide. Two anti-hepatocarcinoma peptides that inhibit the activity of liver cancer Hep3 B cells were obtained, and their peptide sequences were YGRDEISVF and NKSGTYSNDDLSH, respectively. The molecular weight of synthetic peptides YGRDEISVF and NKSGTYSNDDLSH were 1 085.20 u and 1 437.20 u, respectively, and their purity was 98.90% and 98.63%, respectively. The product of hemp seed meal complex enzyme enzymatic hydrolysis contained anti-liver cancer peptides, which provided a theoretical basis for development of anticancer active peptides from hemp seed meal hydrolysate.
