摘 要:目的通过癌症基因组图谱(TCGA)数据库分析prohibitin(PHB)在肾透明细胞癌中的表达及与患者临床特征和预后的关系,为研究肾透明细胞癌的发病机制及治疗方法提供新思路。方法通过TCGA、人类蛋白质图谱(HPA)数据库检索PHB基因在人体正常组织中的表达情况,通过UALCAN数据库分析PHB基因在肾透明细胞癌组织与正常肾组织中的表达水平,并分析PHB表达水平与肾透明细胞癌患者临床特征和预后的关系,利用String数据库预测与PHB基因相互作用的蛋白,并进行基因本位(GO)分析和京都基因与基因组百科全书(KEGG)分析。结果PHB基因在肾透明细胞癌组织中的相对表达量明显低于正常肾脏组织(P﹤0.01)。男性、未分化肾透明细胞癌患者肿瘤组织PHB基因相对表达量分别低于女性、低中高分化患者(P﹤0.05)。PHB低表达肾透明细胞癌患者总生存期短于PHB高表达患者(P﹤0.05)。将交集靶点导入String数据库,得到蛋白质互作网络,共筛选出20个与PHB基因相互作用的蛋白。GO分析结果显示,细胞成分主要集中于细胞内液、细胞质、细胞质膜及线粒体等,生物进程主要集中于细胞内信号转导、细胞死亡调控及细胞凋亡调控等,分子功能主要集中于蛋白结合、ATP结合、蛋白酶激活等;KEGG通路富集分析主要富集于磷脂酰肌醇-3-激酶(PI3K)/蛋白激酶B(AKT)信号通路、免疫反应相关通路等。结论PHB在肾透明细胞癌组织中低表达,PHB表达水平可作为肾透明细胞癌患者诊断、治疗及预后评估的潜在分子标志物。Objective To analyze the expression of prohibitin(PHB)in renal clear cell carcinoma and its relationship with the clinical characteristics and prognosis of patients through The Cancer Genome Atlas(TCGA)database,to provide new ideas for the study of the pathogenesis and treatment of renal clear cell carcinoma.Method The TCGA database and the Human Protein Atlas(HPA)database were used to retrieve the expression of the PHB gene in human normal tissues.The UALCAN database was used to analyze the expression level of PHB gene in renal clear cell carcinoma and normal renal tissue,and to analyze the relationship between the expression level of PHB and the clinical characteristics and prognosis of patients with renal clear cell carcinoma.The String database was used to predict the proteins interacting with PHB gene,and gene ontology(GO)analysis and Kyoto encyclopedia of genes and genomes(KEGG)analysis were carried out.Result The relative expression of PHB gene in renal clear cell carcinoma was significantly lower than that in normal renal tissue(P<0.01).The expression levels of the PHB gene in male and undifferentiated renal clear cell carcinoma patients were lower than those in female and low-medium-highly differentiated patients(P<0.05).The overall survival time of patients with low PHB expression renal clear cell carcinoma was shorter than that of patients with high PHB expression(P<0.05).The intersection targets were introduced into the String database to obtain the protein interaction network,and a total of 20 proteins interacting with PHB gene were screened.Go analysis showed that cell components were mainly concentrated in intracellular fluid,cytoplasm,plasma membrane and mitochondria;biological processes were mainly concentrated in intercellular signal transduction,cell death regulation and apoptosis regulation;and molecular functions were mainly concentrated in protein binding,ATP binding,protease activation,etc.KEGG pathway enrichment analysis mainly concentrated in phosphoinositide 3-kinases(PI3K)/protein kinas
