摘  要:以洋甘菊挥发油为芯材,蛋黄磷脂和胆固醇为壁材,采用乙醇注入-超声法制备洋甘菊挥发油脂质体。通过Box-Behnken响应面法优化洋甘菊挥发油脂质体的配方,观察洋甘菊挥发油脂质体的形态和粒径特征,以及4℃和25℃条件下的稳定性。以大肠杆菌为目标微生物,研究洋甘菊挥发油及挥发油脂质体的抑菌效果。结果表明:洋甘菊挥发油脂质体最优配方为磷脂与胆固醇质量比为5∶1,磷脂与挥发油质量比为4.5∶1,pH为6.8,该条件下制备的脂质体的包封率为91.59%±0.72%,平均粒径为155.37±1.35nm。透射电镜观察脂质体呈球形;稳定性实验说明在4℃条件下脂质体贮存效果好;大肠杆菌抑菌结果表明洋甘菊挥发油脂质体对大肠杆菌具有较好的抑菌活性,并能维持较长的抑菌时间。Chamomile volatile oil was used as the core material,egg yolk phospholipids and cholesterol were selected as wall materials,the chamomile volatile oil liposomes were prepared by ethanol injection-ultrasonic method.The Box-Behnken response surface method was used to optimize formulation of chamomile volatile oil liposomes.Morphology and particle size of the chamomile volatile oil liposomes and their stability at 4℃and 25℃were studied.Escherichia coli.was used as the target microorganism,antibacterial effect of chamomile volatile oil and volatile oil liposomes were tested.The results showed that the optimal formulation of chamomile volatile oil liposomes were:the mass ratio of phospholipids to cholesterol 5∶1,the mass ratio of phospholipids to volatile oil 4.5∶1,and pH value 6.8.The encapsulation efficiency of liposomes prepared under these conditions was 91.59%±0.72%,the average particle size was 155.37±1.35nm.Transmission electron microscopy showed that liposomes were spherical;the stability experiment showed that liposome was good stored at 4℃;the antibacterial results of Escherichia coli showed that chamomile volatile oil liposomes had good antibacterial activity against Escherichia coli.and had longer time antibacterial effects.
