摘 要:目的:评价冬虫夏草提取物及其核苷酸类成分对碱性磷酸酶(ALP)的抑制活性。方法:采用紫外分光光度法测定27个冬虫夏草水提物和5个核苷酸成分的ALP抑制活性,采用分子对接研究5个核苷酸成分在ALP上的结合模式和位点。结果:测定结果表明冬虫夏草对ALP具有较好的抑制活性,不同来源和加工方式样品的ALP抑制活性存在差异。5个核苷酸小分子化合物的ALP抑制活性研究发现,AMP-2Na和CMP-2Na具有相对较好的酶抑制活性。分子对接结果表明氢键和金属-受体相互作用是核苷酸类小分子化合物与ALP结合的重要作用力。结论:研究结果可为虫草类药材治疗疾病及加工提供参考。Objective:To determine the inhibitory activity of water extracts of Cordyceps sinensis and five nucleotides on alkaline phosphatase(ALP).Methods:The ALP inhibitory activity of twenty-seven water extracts of Cordyceps sinensis and five nucleotides were determined by UV-vis spectrophotometry,and the binding mode and site of five nucleotide components on ALP was investigated by molecular docking.Results:The results show that the Cordyceps sinensis has ALP inhibitory activity,and the activities of different processing and different source samples are different.The ALP inhibitory activity of five nucleotide small-molecule compounds found that AMP-2Na and CMP-2Na have relatively good enzyme inhibitory activity.The molecular docking results indicate that hydrogen bonds and metal-receptor interactions are important forces for the binding of nucleotides to ALP.Conclusion:The results of this study can provide references for Cordyceps sinensis medicinal materials in disease treatment and processing.
