摘 要:白灵菇经液体振荡培养得菌丝球,利用Illumina测序平台对其进行转录组测序,获得白灵菇转录组数据,经过质量控制及过滤,共获得26973356条纯净序列,基于RNA-Seq技术和FastQC(v0.11.4)软件对数据进行质量评估,转录组序列共计4.02Gb个碱基,GC含量为53.09%,碱基质量值Q20为98.52%;Q30为95.56%。对测序数据拼接组装后获得32969条Unigene。将获得的Unigene分别与KEGG、Nr、GO和COG等数据库进行比对,共有21808条Unigene得到注释,占总Unigene的66.15%。其中12433条Unigene在GO数据库中得到注释,按照功能分为3个大类和61个亚类;1304条Unigene在KEGG数据库中得到注释,涉及111条代谢通路;21664条Unigene在Nr数据库中获得注释,同源序列主要来自真姬菇;12461条Unigene在COG数据库得到注释,涉及25个不同的功能分类。SSR特征分析中共检测到3014个位点,其中三核苷酸重复1869个,占SSR位点总数的62.01%。本研究构建白灵菇转录组数据库并进行生物信息学分析,为后续开展种质资源鉴定、遗传多样性分析、指纹图谱构建等研究提供科学依据。Pleurotus ostreatus was cultured by liquid shaking to obtain mycelium balls.The transcriptome data of Pleurotus ostreatus were sequenced by Illumina platform,and after quality control and filtration,26973356 pure sequences were obtained.The data were evaluated based on RNA-Seq technology and FastQC(V0.11.4)software,the transcriptome sequence consisted of 4.02 GB bases,with 53.09% GC,98.52% Q20 and 95.56% Q30.After splicing and assembling the sequencing data,32969 unigenes were obtained.Comparing the obtained Unigenes with KEGG,Nr,GO and COG databases,a total of 21,808 Unigenes were annotated,accounting for 66.15% of the total Unigenes.Among them,12,433 Unigenes were annotated in the GO database and divided into 3 categories and 61 sub-categories according to their functions;1,304 Unigenes were annotated in the KEGG database,involving 111 metabolic pathways;21,664 Unigenes were annotated in the Nr database.The annotated homologous sequences were mainly from hypsizygus Marmoreus.There are 12,461 Unigenes annotated in the COG database,involving 25 different functional classifications.A total of 3014 sites were detected in SSR feature analysis,of which 1869 trinucleotide repeats accounted for 62.01% of the total number of SSR sites.In this study,the transcriptome database of Pleurotus ostreatus was constructed and bioinformatics analysis was carried out,so as to provide a scientific basis for subsequent research on germplasm resource identification,genetic diversity analysis and fingerprint construction.
