摘 要:本试验以棉田腐殖质土壤为材料,以醋酸棉酚作为筛选基质,从棉田腐殖质土壤中分离筛选出1株对醋酸棉酚有耐受性的菌株。经生理、生化及16S rDNA基因序列联合分析鉴定,确定该菌株为戊糖片球菌(Pediococcus pentosaceus)。对该菌株的生长曲线、发酵pH、耐热性能、抑菌性能及棉籽粕发酵效果进行测定,结果发现该菌株可在6 h内将发酵体系的pH降低至4左右,且最终pH维持在3.8;在37~45℃菌体存活率大于70%,同时对大肠杆菌、金黄色葡萄球菌具有较强的抑制作用,可清除棉籽粕中87.15%的游离棉酚和22.83%的结合棉酚;与戊糖片球菌标准株ATCC 33316相比,二者对棉籽粕发酵品质的影响无显著差异(P>0.05)。综上所述,本试验所筛菌株在棉籽粕生物发酵脱毒应用方面有极高的应用价值,为研制棉酚脱毒生物制剂提供了优良菌株和科学依据。Using humus soil in cotton field as material and acetate gossypol as screening criteria,a strain with tolerance to gossypol acetate was obtained. It was identified as Pediococcus pentosaceus by physiological and biochemical and 16S rDNA gene sequence analysis. The growth curve,fermentation pH,heat resistant,antibacterial activity and effects on cottonseed meal fermentation were determined. It was found that this strain could reduce the pH of the fermentation system to about 4 within 6 hours,maintained the final pH at 3.8,and the survival rate of the bacteria was greater than 70% in the range of 37 to 45 ℃,and it had inhibitory effect on Escherichia coli and Staphylococcus aureus,and could remove 87.15% free gossypol and 22.83% bound gossypol from fermented cottonseed meal. Compared with the standard strain Pediococcus pentosus ATCC33316,there was no significant difference between them on the fermentation quality in fermented cottonseed meal(P>0.05). The results show that the screened strain has high application value in the biological fermentation and detoxification of cottonseed meal,meanwhile,this study provides excellent strains and scientific basis for the development of gossypol detoxification biological preparation.
