摘  要:文章建立高效液相色谱法(High performance liquid chromatography,HPLC)同时检测食用菌和酵母菌中麦角固醇和V_(D2)的定量方法,以此为基础进行方法学考察。结果表明,优化HPLC检测条件后,麦角固醇和V_(D2)的目标定量峰分别出现在13.7 min和10.6 min。方法学评价显示,食用菌中麦角固醇和V_(D2)定量范围分别为4~350 mg/L和0.15~20.00 mg/L,酵母菌中麦角固醇与V_(D2)的定量范围分别为0.4~36.0 mg/L和0.15~5.00 mg/L,检测线性关系良好,相关系数R^(2)值均大于0.999;食用菌和酵母菌中麦角固醇和V_(D2)的标准平均加标回收率分别为99.2737%(RSD为1.8137%)和108.2293%(RSD为5.8135%),重复性和精密度小于6.5%。通过食用菌与酵母菌中麦角固醇和V_(D2)同时定量检测验证该方法专属性,阐明建立的方法对于食用菌与酵母菌中麦角固醇和V_(D2)同时定量检测具有快速准确的优势。In this paper,a HPLC method for simultaneous determination of ergosterol and V_(D2)in edible fungi and yeast was established.Based on this method,the results showed that the target quantitative peaks of ergosterol and V_(D2) appeared at 13.7 min and 10.6 min,respectively,under optimized detection conditions.The quantitative range of ergosterol and V_(D2) in edible fungi was 4~350 mg/L and 0.15~20 mg/L,respectively.The quantitative range of ergosterol and V_(D2)in yeast was 0.4~36.0 mg/L and V_(D2)0.15~5.00 mg/L,respectively.The detection line had a good relationship.The correlation coefficient R^(2)was greater than 0.999.The average recoveries were 99.2737%(RSD 1.8137%)and 108.2293%(RSD 5.8135%),respectively.The repeatability and precision were less than 6.5%.The specificity of the method was verified by the detection of edible fungi and yeast samples,which indicated that the established method had the advantages of rapid and accurate determination of ergosterol and V_(D2) in edible fungi and yeast.
