马尾连碱乙抑制谷氨酰胺代谢影响胃癌细胞迁移及侵袭的机制

摘　　要：目的 明确马尾连碱乙对胃癌细胞增殖、迁移及侵袭的影响及其参与胃癌细胞能量代谢的方式。方法 两种不同人胃癌SGC-7901、BGC-823细胞系中检测马尾连碱乙对胃癌细胞增殖、迁移及侵袭的影响,同时检测其对不同胃癌细胞能量代谢水平,糖酵解水平及谷氨酰胺代谢水平的影响,最后采用谷氨酰胺代谢酶抑制剂,明确马尾连碱乙参与胃癌能量代谢的作用机制。结果 马尾连碱乙可以显著抑制胃癌细胞的增殖活力,同时降低胃癌细胞的迁移及侵袭能力,给予马尾连碱乙可以明显降低胃癌细胞三磷酸腺苷(ATP)合成能力,不影响细胞乳酸及葡萄糖消耗水平,但可显著抑制胃癌细胞谷氨酰胺代谢能力;马尾连碱乙可以显著抑制谷氨酰胺酶的表达,与谷氨酰胺酶抑制剂联合使用后其对肿瘤细胞增殖及能量代谢的调节能力消失。结论 马尾连碱乙可以抑制谷氨酰胺酶的表达,影响胃癌细胞谷氨酰胺代谢,抑制其能量代谢水平,从而发挥抑制胃癌细胞增殖活力及减少迁移及侵袭的作用。Objective To investigate the effects of caudatine B on the proliferation,migration and invasion of gastric cancer cells and its role in the energy metabolism of gastric cancer cells in vitro.Methods SGC-7901and BGC-823 cell lines were used to detect the effects of equisetine B on the proliferation,migration and invasion of gastric cancer cells,moreover,the effects of equisetine B on the levels of energy metabolism,glycolysis and glutamine metabolism of different gastric cancer cells were detected.In addition the glutamine metabolism inhibitor was used to clarify the action mechanism of equisetine B involved in the energy metabolism of gastric cancer cells.Results The caudatine B could obviously inhibit the proliferation of gastric cancer cells,and decrease the migration and invasion ability as well as ATP synthesis ability of gastric cancer cells,which did not affect the levels of lactic acid and glucose consumption,however,which could significantly inhibit the glutamine metabolism ability of gastric cancer cells.The expression levels of glutaminase were significantly inhibited when caudatine B were combined with glutaminase inhibitor,and its regulatory effects on tumor cells were disappeared.Conclusion The caudatine B can inhibit the expression levels of glutaminase,and affect the glutamine metabolism and energy metabolism of gastric cancer cells,which can inhibit the proliferation ability and decrease the migration and invasion of gastric cancer cells.
