摘 要:以车前草为原料,采用正交试验法,优选大孔树脂提纯车前草中总黄酮的最佳工艺,并以抗坏血酸和芦丁标准品为对照,对车前草总黄酮的抗氧化活性进行研究。结果表明,大孔树脂纯化总黄酮的最佳工艺:AB-8树脂、吸附液pH 6、吸附液质量浓度1.0 mg/mL、解吸液乙醇体积分数90%、上样液体积14 BV、吸附和解吸流速2 BV/h、解吸液体积4 BV。在此工艺条件下,车前草总黄酮产率为3.16%,车前草总黄酮含量由249 mg/g提高到875 mg/g。抗氧化活性方面,纯化后的车前草总黄酮在0.2~2.1 mg/mL范围内,对2,2-联氮-双(3-乙基-苯并噻唑-6-磺酸)二铵盐自由基(ABTS^(+)·)、Fe^(3+)均有不同程度的清除作用,清除率强弱顺序:抗坏血酸>车前草总黄酮>芦丁。表明车前草总黄酮具有一定抗氧化活性,可为车前草的深入开发、产业附加值的提高提供参考数据。Asiatic plantain was used as raw material,and the best process of purifying total flavonoids from Asiatic Plantain with macroporous resin was optimized by orthogonal test.The antioxidant activity of total flavonoids from Asiatic Plantain was studied with ascorbic acid and rutin standard as control.The results showed that the best purification process of total flavonoids by macroporous resin was AB-8 resin,pH of adsorption solution 6,the concentration of adsorption solution 1.0 mg/m L,the ethanol concentration of desorption solution 90%,the volume of loading solution 14 BV,the flow rate of adsorption and desorption 2 BV/h,and the volume of desorption solution 4 BV.Under this condition,the yield of total flavonoids in Asiatic Plantain was 3.16%,and the content of total flavonoids in Asiatic Plantain was increased from 249 mg/g to 875 mg/g.In terms of antioxidant activity,the purified total flavonoids of Asiatic Plantain had scavenging effects on 2,2’-Azinobis-(3-ethylbenzthiazoline-6-sulphonate)free radical(ABTS^(+)·)and Fe^(3+)in the range of 0.2-2.1 mg/mL.The order of scavenging rate was ascorbic acid>total flavonoids of Asiatic Plantain>rutin.The results showed that the total flavonoids of Asiatic Plantain had certain antioxidant activity,which could provide reference data for the further development of Asiatic plantain and the improvement of industrial added value.
