猴头菇成分及提取物抗氧化性和抗癌性研究

摘　　要：为探究药食两用猴头菇中成分以及抗氧化活性和抗癌活性,以猴头菇为原料,对猴头菇中的一般成分、总酚、总黄酮含量进行测定。以80%乙醇和热水分别对猴头菇进行提取,通过对DPPH自由基、ABTS自由基和羟自由基的清除能力评价猴头菇热水提取物和乙醇提取物的抗氧化性;通过MTT试验来分析猴头菇两种溶剂提取物对HepG2和HT29两种癌细胞的抑制作用。结果表明,猴头菇一般成分中粗蛋白和碳水化合物含量分别为24.52%±0.01%和56.56%±0.02%。猴头菇中总酚及总黄酮含量分别为3.43×10^(5)±0.01μg/kg和3.0×10^(5)±0.02μg/kg。乙醇提取物和热水提取物均显示一定的抗氧化和抗癌能力,在测定浓度范围内抗氧化性和抗癌性与提取物浓度呈明显的量效关系。乙醇提取物的抗氧化活性和抗癌活性更优。试验结果表明猴头菇具有良好的抗氧化性和抗癌活性。In order to explore the ingredients,antioxidant activity and anticancer activity of Hericium erinaceus for both medicinal and edible purposes,the general components,total phenols and total flavonoids in Hericium erinaceus were determined by taking Hericium erinaceus as the raw material.The Hericium erinaceus was extracted with 80%ethanol and hot water,respectively,and the antioxidant properties of Hericium erinaceus hot water extract and ethanol extract were evaluated by the scavenging ability of DPPH free radical,ABTS free radical and hydroxyl radical;Analyze the inhibitory effect of two solvent extracts of Hericium erinaceus on HepG2 and HT29 cancer cells.The results showed that the crude protein contents and carbohydrate in the general components of Hericium erinaceus were 24.52%±0.01%and 56.56%±0.02%,respectively.The contents of total phenols and total flavonoids in Hericium erinaceus were 3.43×10^(5)±0.01μg/kg and 3.0×10^(5)±0.02μg/kg,respectively.Both ethanol and hot water extracts showed certain antioxidant and anti-cancer abilities.Within the concentration range,the antioxidant and anti-cancer activities had a significant dose-effect relationship with the extract concentration.The antioxidant activity and anticancer activity of ethanol extract were better.The test results showed that Hericium erinaceus had good antioxidant and anticancer activities.
