摘 要:采用大孔吸附树脂法对黑玉米中的花青素粗提液进行分离纯化,以静态吸附率和解吸率为指标,考察AB-8型、X-5型和DM130型3种大孔吸附树脂对花青素的纯化效果,并通过静态吸附、解吸试验和动态吸附、解吸试验,研究AB-8型大孔吸附树脂纯化黑玉米花青素的最佳条件。结果显示, AB-8型大孔吸附树脂较于其他2种极性树脂,吸附率可达91.3%±1.65%,解吸率可达79.6%±3.44%。AB-8型大孔吸附树脂纯化工艺为静态吸附时间9 h、温度25℃、上样液pH 5;动态吸附和解吸试验中,上样量40 mL、上样流速60 mL/h、解吸液乙醇体积分数50%、解吸液体积65 mL、解吸流速50 mL/h。经过静态试验和动态试验,花青素浓度提高到73.8%。The anthocyanin crude extract from black corn was separated and purified by macroporous adsorption resin method. Taking the static adsorption rate and desorption rate as indicators, the purification effects of AB-8, X-5 and DM130macroporous adsorption resins on anthocyanins were investigated. The optimum condition for the purification of anthocyanins from black corn by AB-8 macroporous adsorption resin was studied. The results showed that the adsorption rate and desorption rate of AB-8 macroporous adsorption resin were 91.3%±1.65% and 79.6%±3.44%, respectively. The purification process of AB-8 macroporous adsorption resin was as follows: the static adsorption time 9 h, the temperature 25 ℃, and the pH of the sample solution 5. In the dynamic adsorption and desorption experiment, the sample loading volume was 40 mL, the sample loading flow rate was 60 mL/h, the ethanol volume fraction of desorption solution was 50%, the desorption solution volume was 65 mL, and the desorption flow rate was 50 mL/h. After static and dynamic tests, the anthocyanin concentration increased to 73.8%.
