摘 要:通过单因素试验优化解淀粉芽孢杆菌BA118的发酵条件,得到BA118的最优发酵条件为接种量2%(V/V),装液量40/250 mL,发酵液初始pH值7.5,发酵温度30℃,转速230 r/min,发酵时间28 h。在单因素试验的基础上,利用Box-behnken试验设计,对培养基成分中的糊精、玉米浆、MgSO_(4)和KH_(2)PO_(4)+K_(2)HPO_(4)(1∶1)4个因素的最优组合进行了定量研究,建立并分析了各因素与菌体密度之间的数学关联模型,获得解淀粉芽孢杆菌BA118的最佳发酵培养基配方为糊精添加量5.50 g/L,玉米浆添加量11.50 g/L,MgSO_(4)添加量3.50 g/L,KH_(2)PO_(4)+K_(2)HPO_(4)(1∶1)添加量6.00 g/L。通过发酵条件和发酵培养基的优化,解淀粉芽孢杆菌BA118的菌体密度达到6.64×10^(9) CFU/mL,提高了28.3倍。The purpose of this research is to study the optimal fermentation conditions and fermentation medium of Bacillus amyloliquefaciens BA118.The fermentation conditions of Bacillus amylolyticus BA118 were optimized by single factor experiment.The optimal fermentation conditions were:culture time 28 h,initial pH 7.5,culture temperature 30℃,rotation speed 230 r/min,inoculation amount 2%(V/V),liquid loading 40/250 mL.On the basis of single factor experiment and Box-behnken experiment design,the optimal combination of dextrin,corn syrup,MgSO_(4) and KH_(2)PO_(4)+K_(2)HPO_(4)(1∶1)in the culture medium components was studied quantitatively.The mathematical correlation model between each factor and cell density was established and analyzed.The optimal fermentation medium formula of Bacillus amylolyticus ba118 was obtained as follows:dextrin 5.50 g/L,corn syrup 11.50 g/L,MgSO_(4)3.50 g/L,KH_(2)PO_(4)+K_(2)HPO_(4)(1∶1)6.00 g/L.The density of Bacillus amylolyticus BA118 reached 6.64×10^(9) CFU/mL and increased by 28.3 times through the optimization of fermentation conditions and fermentation medium.
