摘 要:目的探索蛋白酪氨酸磷酸酶非受体型14(protein tyrosine phosphatase non-receptor type 14,PTPN14)在胰腺癌中的表达水平及其与患者临床病理特征及预后之间的关系。方法通过梳理TCGA数据库中胰腺癌WES、RNA-seq数据及GTEx中正常胰腺组织RNA-seq数据,分析PTPN14在胰腺癌中的突变及比较PTPN14在胰腺癌与正常胰腺组织中表达水平的差异,解析PTPN14表达水平与胰腺癌患者临床病理特征及预后之间的关系。结果(1)PTPN14在胰腺癌中发生错义突变且表达水平高于正常胰腺组织(P<0.05),在基底细胞型(basal)及经典型(classical)胰腺癌中的表达水平也均高于正常胰腺组织(P<0.05)。(2)PTPN14在胰腺癌中的表达水平与肿瘤分化程度(P<0.05)、肿瘤大小(P<0.05)及肿瘤病理类型(P<0.01)相关,与年龄、性别、TNM分期、淋巴结转移及慢性胰腺炎无相关性。(3)PTPN14在胰腺癌中的表达水平与胰腺癌患者总生存期(P<0.05)、无瘤生存期(P<0.05)具有相关性,其表达水平是胰腺癌患者预后不良的独立危险因素(P<0.05)。结论PTPN14在胰腺癌中表达升高,其表达水平可以作为胰腺癌患者预后不良的独立危险因素并且能够预测胰腺癌患者预后状况。Objective To determine the expression profile of protein tyrosine phosphatase non-receptor type 14(PTPN14)in pancreatic adenocarcinoma and its correlation with clinicopathological features and prognosis.Methods Whole exome sequencing data of pancreatic adenocarcinoma were used to analyze the mutation spectrum of PTPN14.RNA-seq data were used to explore the differential expression of PTPN14 in pancreatic adenocarcinoma and normal pancreatic tissues and to elaborate its correlation with clinicopathological features and prognosis.Results 1.Missense mutation of PTPN14 occurred in pancreatic adenocarcinoma and its expression level was higher than normal pancreatic tissue(P<0.05).The expression level of PTPN14 in basal and classical pancreatic adenocarcinoma was also significantly higher than that in normal pancreatic tissue(P<0.05).2.The expression level of PTPN14 in pancreatic adenocarcinoma was correlated with tumor differentiation(P<0.05),tumor size(P<0.05)and tumor pathological type(P<0.01),but not associated with age,gender,TNM stage,lymph node metastasis and chronic pancreatitis.3.The expression level of PTPN14 in pancreatic adenocarcinoma patients was correlated with overall survival(P<0.05)and disease free survival(P<0.05),which was an independent risk factor for prognosis(P<0.05).Conclusion PTPN14 expression was increased in pancreatic adenocarcinoma,and its expression level can be used as an independent risk factor for poor prognosis of pancreatic adenocarcinoma patients and can predict the prognosis status of pancreatic adenocarcinoma patients.
