小鼠肠道乳酸菌定植机制与影响分析

摘  要:乳酸菌被视为一类重要的无芽孢革兰氏阳性菌,可以直接利用碳水化合物实现发酵产酸,在益生菌家族当中扮演着重要的角色,具备调节肠道菌群平衡状态、参与免疫应答以及抑制肠道病原菌生长繁殖等一系列的益生效果。从以往发酵食品当中分离出乳酸菌22株,主要涵盖植物乳杆菌7株、干酪乳杆菌3株、发酵乳杆菌3株、瑞士乳杆菌5株、鼠李糖乳杆菌2株以及保加利亚乳杆菌以及嗜热链球菌各1株等。从以上乳酸菌当中,筛选出6株不同类型的乳杆菌开展动物实验。借助于实时荧光定量PCR技术,探索不同阶段小鼠粪便当中各类乳杆菌的实际数量,分析其在小鼠肠道当中具有的定植能力。由此为工业应用菌株整体功能性的评价及筛选提供相应的参考借鉴依据。Lactic acid bacteria are regarded as an important class of non-spore gram-positive bacteria,which can directly use carbohydrates to ferment and produce acid.They play an important role in the probiotic family,and have the ability to regulate the balance of intestinal flora and participate in immune responses and a series of probiotic effects such as inhibiting the growth and reproduction of intestinal pathogens.22 strains of lactic acid bacteria were isolated from previous fermented foods,mainly including 7 strains of Lactobacillus plantarum,3 strains of Lactobacillus casei,3 strains of Lactobacillus fermentum,5 strains of Lactobacillus helveticus,2 strains of Lactobacillus rhamnosus,Lactobacillus bulgaricus and Lactobacillus bulgaricus 1 strain of Streptococcus thermophilus and so on.From the above lactobacilli,6 different types of lactobacilli were screened for animal experiments.With the help of real-time fluorescence quantitative PCR method,the actual number of various types of lactobacilli in mouse feces at different stages was explored,and their colonization ability in the mouse intestine was analyzed.This provides a reference basis for the overall functional evaluation and screening of industrial strains.
