摘 要:为获得可高产β-苯乙醇的菌株,以β-苯乙醇作为筛选压力进行酵母菌筛选。成功获得一株可耐受4 g/Lβ-苯乙醇浓度的酵母菌YDF-1,可产2.83 g/Lβ-苯乙醇。通过形态学特征和18S rDNA分析鉴定,YDF-1为库德里阿兹威毕赤酵母。并通过单因素和响应面试验优化YDF-1增殖条件,最终优化培养基为葡萄糖100 g/L、胰蛋白胨3 g/L、KH2PO44 g/L、MgSO4·5H2O 0.5 g/L、CuSO40.25 g/L、MnCl20.142 g/L。在以接种量4%,转速250 r/min,30℃的条件下培养20 h,YDF-1可增殖到1.39×109 CFU/mL,相比未优化培养基增加了2.6倍以上。并以此为基础进行酵母增殖转化生产β-苯乙醇,48 h发酵液中β-苯乙醇含量可达到3.27 g/L。To obtain yeast strains that produce highβ-phenylethanol yields,β-phenylethanol was used as the screening pressure.The results showed that the YDF-1 strain tolerating 4 g/L ofβ-phenylethanol was successfully screened and produced 2.82 g/L ofβ-phenylethanol.Based on morphological characteristics and 18S rDNA sequence analysis,YDF-1 was identified as Pichia kudriavzevii.The optimal medium contained 100 g/L glucose,3 g/L tryptone,4 g/L KH_(2)PO_(4),0.5 g/L MgSO_(4)·5H_(2)O,0.25 g/L CuSO4,and 0.142 g/L MnCl_(2).The optimized liquid medium was inoculated at 4%and cultured at 30℃for 20 h at 250 r/min;the bacterial concentration was 1.39×109 CFU/mL,which was more than 2.6 times that of the non optimized medium.This study provides the experimental basis and technical reference to obtainβ-phenylethanol through fermentation.High-density conversion was conducted to produceβ-phenylethanol,and its concentration in 48 h fermentation medium reached 3.27 g/L.
