摘  要:目的 优化直接分光光度法测定保健食品中总黄酮含量的净化溶剂,评估石油醚净化替代苯净化的可行性。方法保健食品样品经乙醇提取、聚酰胺粉吸附后,分别用苯和石油醚净化除杂,甲醇洗脱,在波长360 nm处分光光度法测定其总黄酮的含量。结果 苯净化测定总黄酮回收率为90.3%~94.7%,石油醚净化测定总黄酮回收率为89.5%~93.4%,两者差异无统计学意义(P>0.05)。结论 分光光度法测定总黄酮含量中可以用石油醚替代苯做净化溶剂。Objective To optimize the purification solvent of direct spectrophotometric determination of total flavonoids in health food,the author evaluated the feasibility of replacing benzene with petroleum ether as purification solvent in this study.Methods The health food samples were extracted with ethanol and adsorbed by polyamide powder,purified respectively by benzene or petroleum ether,then eluted with methanol.The content of total flavonoids in the samples was determined by spectrophotometry at wavelength 360 nm.Results The recovery of total flavonoids was in the rage of 90.3%-94.7% purified by benzene and the recovery of total flavonoids was in the rage of 89.5%-93.4% purified by petroleum ether.The difference was not statistically significant(P >0.05).Conclusion Petroleum ether can be used as purification solvent instead of benzene in determination of total flavonoids content by spectrophotometry。
