摘　　要：目的 使用多个生物信息学数据库的数据分析ADAM12 (A Disintegrin And Metalloproteinase12)在骨软组织肉瘤中的表达和预后的相关性及其可能机制。方法 使用UALCAN、GEPIA2.0和Kaplan-Meier Plotter数据库分析ADAM12在多种肿瘤组织中的表达,比较ADAM12在骨软组织肉瘤组织与正常组织中的表达差异并进一步分析其和预后相关性;使用LinkedOmics、Metascape和STRING数据库筛选获得ADAM12共表达基因及相互作用蛋白,富集分析相关信号通路和生物学功能。结果 ADAM12在骨软组织肉瘤等多种肿瘤组织中高表达,且骨软组织肉瘤中ADAM12表达水平高于正常组织(P <0.05),ADAM12高表达的患者总生存期更短(P <0.05);筛选获得RUNX1、ENTPD7、COL5A1等正相关基因,KHK、MORN5、ACBD4等负相关基因;ADAM12相互作用蛋白网络中包括SRC、SDC4、EGF等10种蛋白。结论 ADAM12在骨软组织肉瘤中的高表达与预后负相关,有望作为骨软组织肉瘤潜在的预后生物标志物。Objective To analyze the expression difference of A Disintegrin And Metalloproteinase12(ADAM12) between bone and soft tissue sarcoma and normal tissue,and further investigate its clinical significance and possible mechanism by using bioinformatics databases.Methods UALCAN,GEPIA2.0 and Kaplan-Meier Plotter databases were used to analyze the expression of ADAM12 in various types of cancer as well as the expression difference between bone and soft tissue sarcoma and normal tissue,and clinical significance of ADAM12 in prognosis was further analyzed.LinkedOmics,Metascape and STRING database were used to screen co-expressed genes and protein interaction network of ADAM12 and analyze the signaling pathways and physiological processes involved.Results The expression level of ADAM12 was higher in bone and soft tissue sarcoma compared to normal tissue(P < 0.05).Patients with higher ADAM12 level had poorer OS (P < 0.05).RUNX1,ENTPD7,COL5A1 et al.were positively correlated with ADAM12 expression and KHK,MORN5,ACBD4 et al.were negatively correlated with ADAM12 expression.The protein interation network of ADAM12 contained 10 proteins in total,including SRC,SDC4,EGF et al.Conclusions The expression of ADAM12 is related to the development and poor prognosis of bone and soft tissue sarcoma,it has the potential of being a biomarker for prognosis of bone and soft tissue sarcoma.
