摘 要:目的:分析通江银耳子实体油脂成分及其体外抗氧化活性。方法:采用索氏油脂提取法制备通江银耳子实体油脂,油脂经甲酯化后,采用气相色谱-质谱联用技术进行成分分析,结合NIST数据库进行化合物鉴定,各组分的相对百分含量采用面积归一化法计算。油脂对DPPH自由基的清除能力采用分光光度法测定。结果:基于NIST数据库,从3批通江银耳油脂成分中分离鉴定了12个脂肪酸,各批次中3个主要成分(油酸+棕榈酸+硬脂酸)的含量分别占油脂总含量的92.92%、93.40%和93.08%;在浓度为10.000 mg/mL时,3批银耳子实体油脂对DPPH自由基的清除率分别为19.75%、18.12%、19.23%,EC50值分别为24.07 mg/mL、25.19 mg/mL、24.59 mg/mL,抗氧化作用有明显的剂量依赖性且清除能力低于Vc。结论:棕榈酸和油酸含量可用于通江银耳的鉴别以及油脂定量分析参考指标,通江银耳子实体油脂对DPPH自由基具有一定的清除作用。Objective:In order to analyze the oil components and antioxidant activity of the fruiting body of Tongjiang Tremella fuciformis in vitro.Methods:The oil is prepared by Soxhlet extraction.After methyl esterification,the oil is analyzed and identified by GC-MS.The relative percentage content of each component is calculated by the area normalization method,and the scavenging ability of the oil to DPPH free radical is determined by ultraviolet spectrophotometry.Results:from 3 batches of Tremella fuciformis oil,12 fatty acids are identified based on the NIST database;The contents of the three main components(oleic acid+palmitic acid+stearic acid)in each batch accounted for 92.92%,93.40%and 93.08%of the total oil content,respectively.When the concentration of 3 batches of Tremella fuciformis oil is 10.000 mg/mL,the scavenging rates of DPPH free radicals are 19.75%,18.12%,and 19.23%,and the EC50 values are 24.07 mg/mL,25.19 mg/mL and 24.59 mg/mL,respectively.The antioxidant effect is obviously dosedependent and the scavenging ability is lower than that of Vc.Conclusion:The content of palmitic acid and oleic acid can be used for the identification of Tongjiang Tremella fuciformis and the reference index for quantitative analysis of oil.The fruit body oil of Tongjiang Tremella fuciformis has a scavenging effect on DPPH free radicals.
