摘 要:对10家(贵州省9家、湖南省1家)边销茶生产企业不同年份的15组茶样进行“金花菌”菌落检测,优化检测方法,同时通过DNA提取,ITS序列分析其散囊菌属,确定其优势菌群范围。以同源性最高的比对序列,用MEGA7软件构建系统发育树,了解其亲缘性。结果表明,边销茶中“金花菌”存在较强活性,对于生长环境要求不高,菌落数量较为稳定;优化出一种“金花菌”DNA快速提取方法,极大缩短时间;系统发育树主要分为3个分支,每个分支上遗传距离非常接近。研究可为边销茶中“金花菌”分类鉴定,快速提取其DNA,为后续开展相关研究提供一定的参考。Fifteen samples from 10(9 from Guizhou Province and 1 from Hunan Province)tea producers in different years were tested for"Jinhua fungi"colonies to optimize the detection method,while the DNA extraction and ITS sequence analysis were used to determine the dominant flora.The phylogenetic tree was constructed using MEGA7software to understand their affinities.The results show that the"Jinhua fungi"in the border tea had strong activity,and the growth environment was not high,and the number of colonies was relatively stable.A rapid extraction method was optimized for the DNA extraction of"Jinhua fungi",which greatly shortened the time.The phylogenetic tree shows that"Jinhua fungi"was mainly divided into three branches,and the genetic distances on each branch were very close.This study provided a reference for the classification and identification of the"Jinhua fungi"in the border tea,the rapid extraction of its DNA,and the development of related research.
