摘 要:研究红托竹荪菌托多糖的中试提取工艺及抗氧化能力。采用单因素试验结合正交试验优化红托竹荪菌托多糖中试提取工艺参数,采用总还原力法、DPPH法和羟基自由基法测定多糖的抗氧化能力。优化结果表明,红托竹荪菌托多糖的最佳提取工艺参数为料液比1∶35(kg/L)、提取温度85℃、提取时间2.5 h,重复提取2次,提取液多糖得率可达到18.85%。抗氧化试验结果表明,2.0 mg/mL的多糖对DPPH自由基的清除率可达80.22%,5.0 mg/mL的多糖对OH自由基的清除率可达72.13%,6.0 mg/mL的多糖总还原力约为抗坏血酸的50%。红托竹荪菌托多糖的中试提取工艺合理、可行,所提取多糖具有一定的抗氧化能力。The pilot extraction process and antioxidant capacity of polysaccharides from Dictyophora rubrovalvata volva are studied.The single factor experiment combined with orthogonal experiment is used to optimize the pilot extraction process parameters of polysaccharides from Dictyophora rubrovalvata volva.The total reducing power method,DPPH method and hydroxyl radical method are used to determine the antioxidant capacity of polysaccharides.The optimization results show that the optimal extraction process parameters are as follows:the solid-liquid ratio is 1∶35(kg/L),the extraction temperature is 85℃,the extraction time is 2.5 h,and the extraction is repeated twice.The yield of polysaccharides from the extraction solution could reach 18.85%.The antioxidant experiment results show that the scavenging rate of 2.0 mg/mL polysaccharides on DPPH free radicals can reach 80.22%,the scavenging rate of 5.0 mg/mL polysaccharides on hydroxyl free radicals can reach 72.13%,and the total reducing power of 6.0 mg/mL polysaccharides is about 50%of ascorbic acid.The pilot extraction process of polysaccharides from Dictyophora rubrovvalvata volva is reasonable and feasible,and the extracted polysaccharides have certain antioxidant capacity.
