摘 要:目的:以五味子叶为原料,采用响应面法优化五味子叶多糖(Schisandra chinensis leaves Polysaccharides,SCLP)提取工艺,并研究SCLP抗氧化活性。方法:采用紫外分光光度计法测定SCLP总糖、蛋白质和糖醛酸的含量。在单因素试验基础上,以多糖得率为指标,运用Box-Behnken设计响应面实验优化SCLP的提取工艺。通过测定SCLP对DPPH·、OH·、ABTS^(+)·、和·O_(2)^(-)清除作用考察SCLP的抗氧化活性。结果:五味子叶多糖含总糖(65.21%±2.15%)、蛋白质(5.61%±0.51%)和糖醛酸(16.42%±1.65%)。SCLP最佳提取工艺条件为:液料比为31∶1(mL/g),提取时间为245min,提取温度为81℃,在此条件下多糖得率为7.08%。SCLP具有较强的清除自由基的能力,而且SCLP浓度越高,抗氧化活性越强。结论:本试验得到的工艺条件参数具有可行性,可用于SCLP的提取。SCLP具有良好的抗氧化活性。该研究可为进一步开发利用五味子叶提供科学依据。Objective:In this experiment,using schisandra chinensis leaves as raw material,the technological conditions of SCLP was obtained by using response surface methodology.The antioxidant activity of SCLP was also studied.Methods:The content of total sugar,uronic acid and protein in SCLP was determined by UV.Based on the single factor experiment,the yield of polysaccharides was chosen as the index,and extraction process of SCLP was optimized by Box-Behnken design-response surface methodology.The antioxidant activity of SCLP was studied by free radicals scavenging ability of DPPH,superoxide anion,ABTS and hydroxyl.Results:The total sugar content,the protein content and uronic acid content of SCLP was 65.21%±2.15%,5.61%±0.51%,and 16.42%±1.65%respectively.The optimal extraction conditions were:liquid-to-solid ratio 31∶1(mL/g),245min,81℃.Under these conditions,the yield of polysaccharides was 7.08%.Antioxidant test results showed SCLP had a strong free radicals scavenging ability.The higher the polysaccharide concentration,the stronger the antioxidant activity was.Conclusion:SCLP had good antioxidant activity.The extraction conditions obtained in this experiment was feasible.The study provides a scientific basis for the further development and utilization of Schisandra leaves.
