摘 要:本文研究了一种能节约时间、降低成本且清除自由基能力强的抗氧化肽制备方法。该方法是以鸡胚蛋为原料,利用枯草芽孢杆菌发酵制备鸡胚蛋抗氧化肽。试验用响应面法优化制备,以DPPH·清除率为指标,确定发酵制备工艺的最佳发酵时间、底物浓度和转速,并验证此条件下制备的鸡胚蛋抗氧化肽对自由基的清除能力。结果表明,最佳发酵工艺为:发酵时间为55h,底物浓度为67g/L,转速为193r/min,在此条件下制备的鸡胚蛋抗氧化肽具有较好的清除自由基能力。它对DPPH·清除率高达53.04%,对DPPH·、OH·、O^(2-)·以及ABTS^(+)·的EC_(50)分别为0.94mg/mL、0.26mg/mL、0.22mg/mL、0.012mg/mL。In this paper,an antioxidant peptide preparation which can save time,reduce cost and have strong free radical scavenging ability was studied. The method was used to prepare antioxidant peptides from chicken embryo by bacillus subtilis fermentation. Response surface methodology was used to optimize the preparation. DPPH · clearance rate was taken as the index to determine the optimal fermentation time,substrate concentration and rotation speed as well as the free radical scavenging ability of chicken embryo antioxidant peptide. The results showed that the optimal fermentation conditions were :fermentation time 55h,substrate concentration 67g/L,rotation speed 193r/min. Under these conditions,the prepared chicken embryo antioxidant peptide had better scavenging ability of free radicals. The scavenging rate of DPPH·was 53.04%,and the EC_(50) of DPPH·,OH·,O^(2-)·and ABTS^(+)·were 0.94mg/mL,0.26mg/mL,0.22mg/mL and 0.012mg/mL,respectively.
