摘 要:以沪农灵芝、龙芝2号子实体为原料,分别采用微波提取、碱提醇沉法得到灵芝水溶性和碱溶性多糖.水溶性多糖经体积分数30%、50%、80%乙醇分步沉淀,分别得到不同组分的多糖GLPa、GLPb和GLPc溶液;碱溶性多糖经DEAE-纤维素52柱和葡聚糖凝胶Sephadex G-100柱层析,得到纯化的碱溶性多糖GLPj.采用高效凝胶渗透色谱对不同多糖组分的均一性进行了鉴定,同时测定了相对分子质量.结果表明,低体积分数乙醇沉淀得到的水溶性多糖GLPa相对分子质量达百万级,高体积分数乙醇沉淀得到的水溶性多糖GLPb和GLPc相对分子质量为十万级,碱溶性多糖GLPj相对分子质量小于1万.体外抗氧化活性实验表明,各灵芝多糖均具有还原能力,对羟基自由基及DPPH自由基具有一定的清除作用,灵芝水溶性多糖普遍比碱溶性多糖表现出更好的抗氧化活性,其中GLPa的各项能力较其他多糖均为最佳,抗氧化活性可能与相对分子质量有关.The water-soluble polysaccharides and alkali-soluble polysaccharides of Ganoderma lucidum were obtained by water extraction,alkali extraction and alcohol precipitation from Ganoderma lucidum SH and Longzhi No.2.Water-soluble polysaccharides were precipitated by 30%,50%and 80%ethanol step by step to obtain different fractions of polysaccharides GLPa,GLPb and GLPc.The alkali-soluble polysaccharide GLPj was obtained by DEAE-cellulose 52 column and Sephadex G-100 column chromatography.The molecular weights of each purified polysaccharide component were determined by HPSEC.The results showed that the molecular weight of water-soluble polysaccharide obtained by low concentration ethanol precipitation(GLPa)was one million,water-soluble polysaccharide obtained by high concentration ethanol precipitation(GLPb and GLPc)was 1×105,and alkali-soluble polysaccharide(GLPj)was less than 1×104.In vitro antioxidant activity experiments showed that,all the polysaccharides of Ganoderma lucidum had reducing ability and had certain scavenging effect on hydroxyl radicals and DPPH radicals.Water-soluble polysaccharides generally showed better antioxidant activity than alkali-soluble polysaccharides,among which GLPa had the best ability compared with other polysaccharides,and this may be related to molecular weight.
